摘 要:目的:探讨氧杂蒽酮类的α-倒捻子素(α-Mangostin)、γ-倒捻子素(γ-Mangostin)对U87细胞(Human Malignant Glio-ma Cell)和PC12细胞(Pheochromocytoma Cell)增殖的抑制效应。方法:用MTT方法筛查不同浓度α-倒捻子素、γ-倒捻子素处理U87和PC12细胞48h后的活力效果,Annexin-V/FITC双染后流式细胞仪检测细胞凋亡率。结果:MTT法检测结果显示,α-倒捻子素和γ-倒捻子素能够有效地抑制U87细胞和PC12细胞的增殖,呈剂量依赖性。其中γ-倒捻子素较α-倒捻子素对U87细胞及PC12细胞的增殖抑制作用更强。FCM法检测结果显示,γ-倒捻子素的处理组和对照组比较,PC12细胞的凋亡率差异有统计学意义(P<0.001)。结论:α-倒捻子素、γ-倒捻子素能明显抑制U87细胞和PC12细胞的增殖。γ-倒捻子素能显著性诱发PC12细胞的细胞凋亡。Objective: To investigate the suppression effects of α-/γ-Mangostin on proliferation of U87MG cell and Pheochromocytoma cell.Methods: U87MG cell and Pheochromocytoma cell were cultured and treated with different concentrations of α-/γ-Mangostin in different groups.The proliferation of U87MG cell and Pheochromocytoma cell was detected with MTT assay.Flow cytometry was used to analyze the apoptosis of Pheochromocytoma cell.Results: MTT assay showed that α-/γ-Mangostin significantly inhibited the proliferation of U87MG cell and Pheochromocytoma cell in a dose-dependent fashion.FCM showed that the apoptotic rate of Pheochromocytoma cell was significantly higher in the drug treatment group compared with the control group(P<0.001).Conclusion: α-/γ-Mangostin significantly increased the growth inhibition rate of U87MG cell and Pheochromocytoma cell,at the same time,γ-Mangostin significantly increased the apoptosis ratio of Pheochromocytoma cell.
