摘 要:目的测定不同浓度的胡桃醌对宫颈癌细胞HeLa生长增殖的影响,探讨胡桃醌的抗肿瘤作用。方法体外培养人宫颈癌细胞系HeLa,通过倒置显微镜观察细胞的形态学变化,用MTT试验观察细胞生长增殖状况,流式细胞仪检测细胞周期以及细胞凋亡。结果不同浓度的胡桃醌可导致细胞形态学发生变化。MTT试验显示,与空白对照组比较,不同浓度的胡桃醌对HeLa细胞的增殖数差异均有统计学意义(P<0.05,P<0.01);流式细胞仪检测表明,胡桃醌培养24 h后HeLa细胞出现G2/M期阻滞,其细胞凋亡率明显增加,与对照组比较,不同浓度的胡桃醌差异均有统计学意义(P<0.05,P<0.01)。Caspase-3活性检测结果显示,与空白对照组比较,不同浓度的胡桃醌可使Caspase-3的活性增加,差异有统计学意义(P<0.05,P<0.01);以上全部结果具有剂量依赖性。结论不同浓度的胡桃醌可以抑制HeLa的增殖,且具有剂量依赖性。Objective To detect the effect of juglone at different concentrations on growth and proliferation of HeLa cells. Methods The human HeLa cell line was cultivated in vitro. The changes of morphology were observed by inverted microscope ; the propagation was detected by MTF and the cell cycle and cell apopto- sis were detected by flow cytometer. Results Cytomorphology trial showed that juglone at different concentrations could lead to changes of cellular morphology. MTY showed that there was significant difference in juglone at different concentrations compared with blank control group ( P 〈 0.05, P 〈 0.01 ) ; the flow cytome- ter showed that after 24 hours, the G2/M phase was appeared in HeLa cells and the apoptosis rate was increased. Compared with control group, there was signifi- cant difference in juglone at different concentrations ( P 〈 0.05, P 〈 0.01 ) ; the Caspase-3 activity assay showed that juglone at different concentrations could in- crease the activity of Caspase-3, and which was significantly different from the control group ( P 〈 0.05, P 〈 0.01 ) ; all the results above had dose-dependent. Conclusion Juglone at different concentrations can inhibit the proliferation of HeLa cells, which have dose-dependent, and expected to become the new anti- tumor therapeutic targets.
