摘 要:在Cu2+与色氨酸物质的量浓度比为0至40范围内,文章用F-2500荧光仪分别对0.1 MPa和60 MPa条件下酪氨酸三维荧光进行了测量,并对三维荧光光谱的平均值、标准差和重心进行了提取。结果表明:色氨酸的荧光强度随着压力的增加而增加,当压力由0.1 MPa变化为60 MPa时,纯色氨酸的最大值增加了3.4%。Cu2+对色氨酸具有猝灭作用,而压力的增加使得色氨酸的猝灭作用减弱,当Cu2+与色氨酸的物质的量浓度比由0增加到40,当压力为0.1 MPa的时候,平均值和标准差分别降低了2.1%和2.9%;但当压力为60 MPa的时候,平均值和标准差分别降低了1.5%、1.5%。压力从0.1 MPa增加到60 MPa的时候,色氨酸重心纵坐标增加了2 nm。The three - dimensional fluorescence spectra of tryptophan solution with different concentrations of copper ion under 0. 1 MPa and 60 MPa were detected through F - 2500 made in Japan. And the characteristic param- eters " mean" ," deviation" and" barycenter" were extracted based on the principle of mathematical statistics. The re- suits showed that the "mean" increased with the increase of pressure, and the maximum fluorescence intensity in- creased by 3.4% when the pressure changed from 0.1 MPa to 60 MPa. The fluorescence of tryptophan aqueous solu- tion was quenched obviously due to complex formation with Cu2+. The quenching weakened when the pressure in- creased. The "mean" and "deviation" decreased by 2.1% and 2.9% under 0.1MPa when the concentration ratio ofCu2+ and tryptophan ted the fluorescence changed from 0 to 40, while 1.5% and 1.5% under 60 MPa. It was observed that pressure shif- to red direction through the comparing the ordinate of barycenter.
