环孢素预处理对同种带瓣主动脉移植后钙含量的影响及意义

摘　　要：目的探讨环孢素预处理对同种异体带瓣主动脉移植后钙含量的改变及其意义。方法 4周龄的白兔50只(取带瓣动脉)为供体,体重0.30～0.35 kg;8周龄白兔50只,体重3.00～3.50 kg,随机分2组为受体,25只(环孢素预处理移植带瓣主动脉)实验组,25只(环孢素预不处理移植带瓣主动脉)对照组,分别于术后2、4、8周取出血管测定CD25和CD40的表达,原子吸收光谱法测定组织钙含量。结果①对照组CD25和CD40表达水平的高峰在移植后2周,此后逐渐回落,实验组在移植后2～4周的表达明显低于对照组(P<0.01);②实验组在2、4、8周3个不同时间点的钙含量明显低于对照组(P<0.01)。结论移植后使用环孢素抑制了早期的免疫排斥反应,降低了同种带瓣血管的钙化程度。Objective To explore the effect and significance of cyclosporine pretreatment on calcium content after aorta homograft with valve. Methods 50 China white rabbits aged 4 weeks and weighed 0.30-0.35 kg were enrolled as donors of aorta with valve. 50 rabbits of 8 weeks with weight of 3.00 - 3.50 kg were randomly divided into experimental group that were pretreated with cyclosporine and the control group,each with 25 rabbits. Aorta of the two groups was taken out 2,4, and 8 weeks after graft to determine the express level of CD25 and CD40 and atomic absorption spectrometry calcium. Results CD25 and CD40 expression level in the control group reached the peak in 2 weeks after transplantation, then gradually came hack, and were significantly lowed than those in the experimental group at the time point of 2 to 4 weeks after graft. The calcium content of the experimental group were significantly lower than that of the control group at all three time point of 2,4,8 weeks after graft （ P 〈 0.01 ）. Conclusion Cyclosporine can inhibit early immunological rejection and decrease the calcium degree of homograft blood vessel.
