高效液相色谱法测定益气消肿合剂中葛根素含量

摘　　要：目的:利用高效液相色谱法以葛根素为指标对益气消肿合剂进行定量分析,建立益气消肿合剂的含量测定方法。方法:采取反相高效液相色谱法,利用C18色谱柱,以甲醇-水(25∶75)为流动相,在250 nm波长下测定益气消肿合剂中葛根素含量。结果:在上述测定条件下,葛根素在210.5 ng～4 210 ng范围内线性关系良好。测定方法重复性良好,RSD=1.71%(n=6),益气消肿合剂中葛根素的平均回收率%=102.00(n=5),RSD=0.54%。结论:该方法准确性、灵敏度、重复性高,简便,准确,易操作,可作为益气消肿合剂的质量控制方法。Objective ： To do quantitative analysis for Mistura of Yi-qi Xiao-zhong with Puerarin as an index and high performance liquid chromatography as the method, and set up the assay method for Mistura of Yi-qi Xiao- zhong. Methods：Puerarin content in Mistura of Yi-qi Xiao-zhong was determined in 250nm wave-length with re- versed phase high-performance liquid chromatography （RP-HPLC）, C18 chromatographic column and methanol - water （25：75 ） as mobile phase. Results： In the condition J of linear relationship between 210.5ng and 4210ng range,the repeatability of puerarin for the determination is good,RSD = 1.71% （n =6） ,and the average recovery percent of Puerarin in Mistura of Yi-qi Xiao-zhong was 102 （n = 5 ）, RSD = 0. 54%. Conclusion：This method is ac- curate, sensitive, reproducible, simple and easy to operate, and can be used as the quality control method for Mistu- ra of Yi-qi Xiao-zhong.
