摘 要:以铁线蕨(Adiantum capillus-veneris L.)不同世代植株为材料,对铁线蕨总DNA不同提取方法进行比较,并通过检测体系进行分析.结果表明:结构简单的配子体为材料的DNA的纯度明显高于孢子体,但是产量也明显低于孢子体;对于配子体而言,CTAB法、SDS法、高盐低pH法、尿素提取法、试剂盒法都能提取较为纯净的总DNA,但经过DNA完整性检测体系筛选后,改良CTAB法结果最优;对于含大量次生代谢产物的孢子体植株而言,改良CTAB法得到的总DNA纯度明显高于其它,通过后续的DNA完整性检测得到与配子体相同结论.Different whole DNA extraction protocols were compared using the two different materials of A.capillus-veneris L.of different life cycles.And the results of different measuring systems were analysed.The results indicated that: compared to the sporophytes,the purity rate of the gametophytes was apparently higher but the output was lower;To the gametophyte,these nine methods could extract much pure DNA products;But after the screen of the detection PCR,the methods of CTAB,SDS and the Kit were the best To the sporophyte,the whole DNA purity obtained by the modified CTAB DNA extraction method was obviously better than others,the detection PCR also got the same results.
