摘 要:目的探讨苏木、川芎含药血清对常氧、低氧环境下培养的人高转移性巨细胞肺癌(PG)细胞增殖的影响,筛选合适的含药血清浓度,明确最佳作用时间。方法制备苏木、川芎含药血清,分别用含药血清干预常氧、低氧环境下培养的PG细胞,按照含药血清浓度不同分为6组,同时设对照组。采用噻唑蓝(MTT)法观察苏木、川芎含药血清对PG细胞增殖的影响。结果在常氧环境下,苏木含药血清各浓度之间(除浓度为0.625%)、川芎含药血清各浓度之间(除浓度为1.25%)差异均有统计学意义(P<0.05);在低氧环境下,苏木、川芎含药血清各浓度之间差异均没有统计学意义(P>0.05)。结论苏木、川芎含药血清在常氧环境下作用24 h时能抑制PG细胞的增殖,并呈浓度依赖,浓度越高,抑制作用越明显,但在低氧环境下则促进PG细胞增殖;随着苏木、川芎含药血清作用时间的延长(48、72 h),在常氧、低氧环境下其含药血清均有促进PG细胞增殖的作用(72 h时作用最明显),并且呈浓度依赖,浓度越高,越有利于PG细胞增殖(苏木、川芎浓度为20%的时候作用最明显)。低氧环境更有利于苏木、川芎含药血清促进PG细胞增殖。Objective To investigate the influences of medicinal serum of Sumu (Lignurn Sappan) and Chuanxiong (Rhizoma Ligutici) on the proliferation of human highly metastasis PG cells cultured in normoxia and hypoxia conditions, screen the suitable concentration of medicinal serum and determine the best acting time. Methods The medicinal serum samples of Sumu and Chuanxiong were prepared and used to intervene PG cells cultured in normoxia and hypoxia conditions. The samples were then divided into 6 groups according to the concentration of medicinal serum and a control group was set. The influences of medicinal serum of Sumu and Chuanxiong on the proliferation of PG cells were observed by using MTr method. Results In normoxia condition, the differences among different concentration of medicinal serum of Sumu ( except of 0.625% ) and medicinal serum of Chuanxiong ( except of 1.25% ) had statistical significance (P 〈 0. 05 ). In hypoxia condition, concentration of medicinal serum of Sumu and medicinal serum differences among different of Chuanxiong had no statistical significance (P 〉 0.05). Conclusion The medicinal serum of Sumu or Chuanxiong can inhibit PG cell proliferation in normoxia condition after intervening for 24 hours and shows a dose-dependence. The higher the concentration of the serum is, the more obvious its inhibitory effect is. But in hypoxia condition, the medicinal serum can improve PG cell proliferation. With its action time increasing (48 h and 72 h ) , the medicinal serum has the action of promoting PG cell proliferation ( being the most significant at 72 h) in normoxia and hypoxia conditions, and shows a dose-dependence. The higher the concentration of the serum is, the more significant PG cell proliferation is (being the most obvious in concentration of 20% ). The condition of hypoxia is more beneficial to the proliferation of PG cells promoted by the medicinal serum of Sumu or Chuanxiong.
