摘 要:目的:RP-HPLC法和氨基酸分析仪(amino acid analyzer,AAA)法测定氨基酸含量的比较。方法:采用高效液相色谱仪和氨基酸分析仪测定肠外营养注射液(25)中18种氨基酸的含量。采用C18柱和异硫氰酸苯酯进行柱前衍生化,加入二硫代二丙酸作为保护剂,同时测定半胱氨酸;AAA法采用离子交换色谱柱和茚三酮柱后衍生化进行,不需加入保护剂,直接测定半胱氨酸。结果:RP-HPLC法和AAA法的重复性分别小于1.1%和3.1%,2种测定方法的相对偏差在0.19%~8.2%之间。结论:AAA法更适于肠外营养注射液(25)种18中氨基酸的测定。Objective:To compare the RP-HPLC and amino acid analyzer(AAA) methods for determining concentrations of amino acids.Method:Eighteen kinds of amino acids in total parenteral nutrition injection(25) were measured by an HPLC and amino acid analyzer.One C18 column was used for HPLC method with phenyl isothiocyanate precolumn derivation,dithio-dipropanoic acid was added as protective agent and cysteine was determined in the meanwhile;an ion exchange chromatography column was used for AAA method with ninhydrin postcolumn derivation,no protective agent was used and cysteine was determined directly.Result:The repeatability of RP-HPLC and AAA methods were less than 1.1% and 3.1%,respectively.The standard deviation of two methods ranged from 0.19%-8.2%.Conclusion:AAA method is more suitable for determining the concentrations of amino acids than HPLC method.
