摘  要:目的体外观察白芷活性提取物对人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HUVECs)生物学特性的影响,初步探讨其促进创面愈合的可能机制。方法按照随机数字表法分5×10-2g/L、5×10-3g/L、5×10-4g/L、5×10-5g/L白芷活性提取物组,以0.25%fcs/DMEM为对照组。采用噻唑蓝比色法检测不同浓度白芷活性提取物对体外培养的HUVECs增殖作用的影响。通过实时荧光定量聚合酶链式反应分别检测白芷活性提取物最适浓度培养条件下HUVECs的细胞周期相关基因cyclin D1、凋亡基因Caspase3、VEGF的mRNA表达情况。结果白芷活性提取物在5×10-3~5×10-2g/L浓度间可促进HUVECs增殖,呈剂量依赖性,吸光度值分别为(0.335±0.027、0.359±0.033),与对照组(0.306±0.023)相比差异具有统计学意义(t值分别为1.84、3.03,P值均小于0.05),其中在5×10-2g/L浓度时测得吸光度值最高,促增殖作用最为显著(t=3.03,P<0.01),为白芷活性提取物促进HUVECs增殖的最适浓度。白芷活性提取物组cyclin D1 mRNA相对表达量(0.446±0.054),低于DMEM对照组(1.000±0.150),差异有统计学意义(t=5.508,P=0.00028<0.01),白芷活性提取物组Caspase3 mRNA相对表达量(0.600±0.052),低于DMEM对照组(1.000±0.165),差异有统计学意义(t=4.67,P=0.0047,<0.01),白芷活性提取物组VEGF mRNA相对表达量(0.839±0.060),低于DMEM对照组(1.000±0.169),差异无统计学意义(t=1.709,P=0.0628,>0.05)。结论白芷活性提取物能显著促进HUVECs增殖,下调cyclin D1的表达加快细胞周期进程,同时下调Caspase3 mRNA的表达抑制细胞凋亡,且可能与白芷促进创面愈合的机制有关。Objective To observe the effects of Angelica dahurica extract on the biological characteristics of human umbilical vein endothelial cells (HUVECs) in vitro and to preliminarily explore the possible therapeutic mechanism on inducing wound healing. Methods Cultured human umbilical vein endothelial cells ( HUVECs ) , different concentrations of the Angelica dahuriea extracts ware added to the medium. HUVECs were treated with different concentration of Angelica dahurica extracts. The proliferation activity of HUVECs was measured by using thiazole blue (MTT) colorimetric assay. The cyclin D1, Caspase 3 and VEGF mRNA levels of treated HUVECs were detected by real-time polymerase chain reaction (PCR) techniques. Results The Angelica dahurica extracts ( at the concentration from 5 × 10 ^-3 g/L - 5 × 10 ^-2 g/L) can significantly enhance HUVECs proliferation. The effect of enhancing proliferation was the most significantly at the concentration of 5 × 10 ^-2 g/L ( t = 3.03, P 〈 0.01 ). Real-Time PCR revealed that compared with control group, the cyclin D1 and Caspase3 mRNA level in the concentration of 5 × 10^- 3 g/L Angelica dahurica extract group was significantly down-regnlated, VEGF mRNA level no significantly different. Conclusions Angelica dahurica extract can notably promote the proliferation of HUVECs, accelerate the cell cycle of HUVECs and down-regulate the mRNA expression of Caspase 3. These effects may related with the mechanism of Angelica dahurica extract on enhancing the process of wound healing.
