摘  要:目的研究回转模拟微重力(simulated microgravity,SMG)条件下成骨生长肽(OGP10-14)对MC3T3-E1成骨样细胞增殖与分化的影响。方法利用回转器模拟失重,MTT法检测OGP10-14对回转24 h,48h,72 h MC3T3-E1细胞增殖的效应,并观察培养液中碱性磷酸酶(alkaline phosphatase,ALP)、骨桥素(osteopontin,OPN)活性变化。结果回转模拟微重力条件下,10-8mol/L OGP10-14能够显著促进成骨细胞系MC3T3-E1的增殖(P<0.01)并促进成骨细胞系MC3T3-E1的ALP和OPN活性。结论回转模拟微重力条件下,OGP10-14对成骨细胞系MC3T3-E1的增殖与分化具有促进作用。Objective To study the effect of osteogenic growth peptide ( OGPl0-14 ) on the differentiation and proliferation of osteoblast MC3T3-E1 under the conditions of simulated microgravity (SMG). Methods Under SMG 24 h, 48 h and 72 h, the effectiveness of the proliferation and differentiation of osteoblasts MC3T3-E1 were examined by MTT. Activity changes of ALP and OPN in the cell culture medium was observed after treated with OGP10-14. Results Under SMG, 10-s mol/L OGP10-14 were able to significantly promote the proliferation of osteoblasts MC3T3-EI(P 〈0.01 ) and enhanced the ALP and OPN activity of osteoblasts MC3T3-E1. Conclusion Under SMG, OGP10-14 facilitates the proliferation and differentiation of osteoblasts MC3T3-E1.
