摘 要:目的:研究外源性核苷酸(exogenous nucleotides,EN)对乙醇发育毒性的拮抗作用。方法:将C57BL/6J孕鼠随机分为空白对照组、乙醇对照组和4个EN干预组(EN的质量分数分别为0.01%、0.04%、0.16%、0.64%);孕6~15 d时空白对照组以每千克体重5 g蒸馏水灌胃,乙醇对照组和EN干预组以相同剂量的乙醇灌胃,建立乙醇发育毒性动物模型;整个实验周期内,2个对照组动物喂饲普通小鼠饲料,4个EN组动物喂饲含有相应剂量核苷酸的干预饲料。孕第18天收集母鼠血液,检测血清中的超氧化物歧化酶(superoxide dismutase,SOD)活力、丙二醛(malondialdehyde,MDA)含量和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活力;称量活胎和胎盘重量,观察胚胎毒性;并进行活胎骨骼和内脏畸形检查。结果:与乙醇对照组相比,核苷酸干预组平均活胎重量、胎盘重量均有所改善;孕鼠血清中SOD活力和GSH-Px活力明显提高(P<0.05),MDA含量显著降低(P<0.01);胎鼠活胎率显著增加,吸收胎率显著下降(P<0.01);胎鼠四肢骨和尾椎骨骨化数目均明显增加(P<0.01)、枕骨和胸骨骨化异常发生率有明显下降(P<0.05),其中以0.04%EN组最为明显。结论:孕期补充EN对乙醇的发育毒性具有一定的拮抗作用。Objective: To investigate the preventive developmental toxicity of alcohol. Methods: C57BL/6J effect of exogenous nucleotide (EN) on the pregnant mice were divided into 6 groups ran- domly: the control group, the alcohol group, the low (0. 01% EN), middle (0. 04% EN), high (0.16%EN)and higher (0.64%EN) intervention groups. From the 6th gestational days (GD) to the 15th GD, pregnant animals except those in the control group were administrated with 5 g (per kg body weight) alcohol intragastrically. Normal mouse forage was provided to the animals in the control group and alcohol group, while forage containing different quantities of EN was provided to the animals in the intervention groups. On GD 18, all the dams were killed, their blood samples were collected for further analysis, and fetal developmental indexes were observed. Results: Compared with the animals in the alcohol group, offspring' s body weight and placenta weight of EN intervention groups improved a lot; superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity increased significantly (P 〈 0.05), malondialdehyde (MDA) levels decreased significantly (P 〈0.01 ); the live birth rate of ani- mals in the interventions groups has increased significantly and the absorbed embryo rate of them has decreased significantly ( P 〈 0.01 ) ; the forepaw p ossification point increased significantly ( P 〈 0.01 ) decreased significantly in 0.04% EN group ( P 〈 0. halanges, hindpaw phalanges and caudal vertebrae ; the anomalies of both occipital and sternum have 05 ). Conclusion: EN has a significant protective effect on the developmental toxicity of alcohol.
