摘 要:以冬瓜子叶节为试材.探讨培养条件、不同基因型及诱导、伸长和生根培养基不同激素类型与配比情况.初步建立了冬瓜组培再生体系。结果表明:种子剥皮消毒后.在纸桥培养基暗培养发芽率较高,且节约成本:以暗培养5d、见光培养1d刚转为淡绿色的闭合子叶,其不定芽诱导率最高,约75%;以13个不同基因型冬瓜子叶节为外植体.接种至不同激素浓度组合的培养基上,筛选出分化率较高的品种B214,其不定芽再生率可达到79-2%.诱导分化培养基为MS+4.0mg/L6一BA+0.4mg/LNAA,伸长培养基为MS+I.0mg/L6-BA+0.5mg/LNAA.生根培养基为1/2MS+0.5mg/LIAA;当冬瓜组培植株高4-5cm时,炼苗移栽,其移栽成活率在60%以上。Cotyledonary nodes of Benincasah. hispida were used for tissue culture to establish a high-frequency regeneration system, and the effect of culture condition, different genotypes of Benincasah. hispida, hormones types and combinations were groped on the adventitious buds. The results showed that germination percentage of seeds was the highest and the cost was also saved on paper-bridge culture medium; the highest inducing frequency of adventitious buds was found when cotyledons cultured 5 days under dark and 1 day under light were closing and greenish; Cotyledonary nodes of 13 genotypes as explants were cultured on inducing mediums supplemented with different hormones concentration combinations. The resuhs indicated that B214 were the highest on the medium (MS+4.0 mg/L6-BA+0.4 mg/L NAA)and differentiation percentage was up to 79.2%. The appropriate elongation and radication media were MS +1.0 mg/L 6-BA +0.5 mg/L NAA and 1/2 MS +0.5 mg/L IAA respectedly. Tissue culture seedlings of Benincasah. hispida with the height of 4-5 cm got through seedling adaptation, resulting in a survival rate of up to 60%.
