大鼠骨髓间充质干细胞促进超比例任意皮瓣成活的初步实验研究

摘　　要：目的观察大鼠骨髓间充质干细胞(BMSCs)对超比例任意皮瓣成活的影响。方法采用全骨髓贴壁培养法获得大鼠骨髓间充质干细胞,通过成骨、成脂肪诱导分化以及流式细胞仪分析其表面标记鉴定BMSCs特征;在SD大鼠背部形成超比例任意皮瓣模型,将BMSCs皮瓣内注射治疗,与生理盐水作对照。观察皮瓣大体情况,计算皮瓣成活率。结果全骨髓贴壁培养法获得的BMSCs经诱导后可分别向成骨、成脂转化;流式分析表面标志分子高表达CD90、CD29,低表达CD34、CD45。皮瓣术后2周,皮瓣成活与坏死部分界限清楚,BMSCs治疗组皮瓣成活率明显高于生理盐水组。结论 BMSCs可明显增加超比例任意皮瓣成活面积,其在临床治疗中有潜在的应用和研究价值。Objective To observe the effect of hyper-proportion random flap Survive with BMSCs.Methods Culture was performed with adhering method;characteristic of BMSCs was identified through multipotent differentiation and surface marker assay.The model of hyper-proportion flap was obtained on SD rat＇s back,TO contrast with the treated group and the control group,which was treated by subcutaneouly injected with BMSCs,the other group with injecting saline.Then the survival rate of the flap was measured.Results BMSCs can be separated and proliferated effectively by the whole bone marrow adherence culture,The surface marker assay was higher expressive CD90,CD29,and lower expressive CD34,CD45 by using flow analyzer.After two weeks,the line was clearly between survival and necrotic flap.The survival rate of the treated group was significantly higher than the control group.Conclusions BMSCs can increase survival area of random skin flap,and has potential application and research value in clinical work.
