摘 要:目的:研究雷帕霉素(rapamycin,RAPA)对未成熟树突状细胞(immature dendritic cell,imDC)RelB表达的影响,为回输RAPA处理的imDC(RAPA-imDC)诱导移植耐受的机制研究提供实验依据。方法:分离诱导imDC、RAPA-imDC、成熟树突状细胞(mature dendritic cell,mDC),光镜和电镜观察其形态,流式细胞仪检测其CD80、CD86、MHC-Ⅱ表达,用免疫荧光法检测其RelB表达。结果:①电镜结果显示imDC与RAPA-imDC形态相似,呈未成熟状态,树突少、粗短;mDC树突较多,细长。②mDC表面CD80、CD86、MHC-Ⅱ表达显著高于imDC和RAPA-imDC(P<0.05);而imDC的表达也明显高于RAPA-imDC(P<0.05)。③免疫荧光法检测结果显示imDC、RAPA-imDC RelB表达量无显著变化,而mDC表达量高。结论:雷帕霉素能降低imDC表面的CD80、CD86、MHC-Ⅱ表达,但对其RelB表达没有明显影响。Objective: To study the effect of rapamycin (RAPA) on RelB expression of immature dendritic cell (imDC), to provide an experimental basis for understanding the transplantation tolerance mechanism induced by the RAPA-conditioned imDC (RAPA- imDC). Methods: imDC, mDC and RAPA-imDC were isoltated and culctured. Their morphology was observed by light and electron microscopy. Their CD80,CD86,MHC- Ⅱ were detected by flow cytometry.Their RelB expressions were measured by immunofluore-seence and Western blotting. Results: The morphology of RAPA-imDC was similar to the imDC. Their dendrons were few, more coarse and shorter. The dendrons of the mDC were more, longer and thinner. The expression of surface molecules CD80, CD86, MHC-Ⅱ on mDC was the highest, and they were higher on imDC than on RAPA-imDC. RelB expression was higher on mDC.However it was low on imDC and RAPA-imDC without statistical difference. Conclusion: Rapamycin can reduce CD80,CD86,MHC-Ⅱ moleculars on imDC, but has no effect on its RelB expression.
