摘 要:目的优化大孔吸附树脂法纯化广金钱草总黄酮工艺。方法采用静态与动态吸附-解吸附方法对广金钱草总黄酮进行纯化,以吸附率与洗脱率等指标优化工艺,利用紫外-可见分光光度计测量广金钱草总黄酮含量。结果最佳工艺:HPD100大孔树脂、上样液pH4、上样浓度4 mg/mL、洗脱溶剂60%乙醇、洗脱速度1.8BV/h、洗脱体积6BV、最佳吸附量16mg/g、树脂柱径高比1:6,纯化后总黄酮含量高达90.85%。结论HPD100大孔树脂吸附分离工艺能有效地对广金钱草总黄酮进行富集纯化。Objective Purify the total flavonoids from Desmodium styracifolium (Osbeck) Merr. with macro- porous resin. Methods The optimal techniques of separation and purification of flavonoid were obtained by static adsorption and elution tests, and the separation techniques were evaluated by measuring the concentration of total flavonoids from D. styracifolium with ultraviolet spectropho tometer. Results The optimum technology was as fol- lows : the HPD-100 macroporous resin , concentration 4mg/mL, pH 4 and the concentration of elution was 60% al- cohol, eluting velocity 1.8BV/h ,elution volume of 6BV, the proportion of diameter and height were 1: 6. After purified by HPD-100, the extract of total flavonoids could be up to 90.85% . Conclusion HPD100 macroporous resin adsorption separation process can be effectively applied to purify total flavonoids from Desmodium styracifolium.
