摘 要:目的探讨磷脂氢谷胱甘肽过氧化物酶(PHGPx)在p,p’-DDE对大鼠睾丸生精细胞脂质过氧化及诱导生精细胞凋亡中的作用。方法将20只健康清洁级50日龄雄性SD大鼠按体重随机分为4组,分别为对照(玉米油)组及低(20 mg/kg)、中(60 mg/kg)、高(100 mg/kg)剂量p,p’-DDE染毒组,每组5只。采用腹腔注射方式染毒,注射容积为10ml/kg,每隔1 d染毒1次,连续进行10 d。测定血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活力和丙二醛(MDA)的含量。采用Western blotting方法和检测大鼠睾丸组织PHGPx蛋白相对表达情况,采用实时荧光定量PCR(FQ-PCR)测定大鼠睾丸组织PHGPx及B细胞淋巴瘤-2相关X蛋白(Bcl-2 associated protein X,Bax)、细胞色素C(Cyt C)、凋亡蛋白酶活化因子-1(Apaf-1)、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)mRNA的表达水平。结果与对照组相比,60 mg/kg p,p’-DDE染毒组血清SOD、GSH-Px活力和睾丸组织PHGPx蛋白表达水平均降低,100 mg/kg p,p’-DDE染毒组大鼠睾丸组织中PHGPx、Cyt C、Apaf-1 mRNA表达水平和60 mg/kg p,p’-DDE染毒组Caspase-3 mRNA表达水平较高,60 mg/kg、100 mg/kg p,p’-DDE染毒组Bax mRNA表达水平均升高,差异有统计学意义(P<0.05)。结论PHGPx在p,p’-DDE所致男(雄)性生殖功能紊乱中起拮抗作用。Objective To study the role of PHGPx in the lipid peroxidation and apoptosis of testicular germ cells initiated by p ,p' -DDE. Methods A total of 20 male SD rats were randomly divided into four groups (three exposure groups and one control group), five rats in each group, and the rats were given different doses ofp ,p '-DDE(0,20,60,100 mg/kg respectively) in corn oil every other day by intraperitoneal injection for ten days. The rats were sacrificed after ten days of treatment,and the blood was collected, the levels of SOD, MDA, GSH-Px in serum were measured with the assay kits; Western blotting was used to detect the protein of PHGPx; Real-time PCR was used to detect PHGPx, Bax, Cyt C, Apaf-1 and caspase-3 mRNA. Results Compared with the control group, the activities of SOD and the GSH-Px level were reduced in group of 60 mg/kg p ,p'-DDE, the PHGPx protein expression level was reduced in the moderate p ,p'-DDE dose group,meanwhile,the mRNA levels of PHGPx, Cyt C and Apaf-1 in the high p ,p'-DDE dose groups were higher,the mRNA levels of Casepase-3 were elevated in group of 60 mg/kg p ,p' -DDE, and the mRNA levels of Bax in groups of 60,100 mg/kg p ,p' -DDE were increased, the differences were statistically significant(P〈0.05). Conclusion PHGPx has antagonistic action in the reproductive dysfunction caused by p ,p'-DDE in pubescent male rats.
