摘 要:目的探讨围产期低剂量氯化三丁基锡(TBTCl)暴露对子代雄性小鼠青春期精子质量的影响。方法将20只妊娠6 d的SPF级昆明小鼠随机分为4组,分别为100、10、1μg/kg TBTCl染毒组和对照(玉米油)组,每组5只。从妊娠第6天开始,于每天同一时间经口灌胃染毒,染毒容量为1 ml/kg,至哺乳期结束(出生后21 d)。于出生后28 d开始观察包皮分离的日龄。出生后第49天,取附睾做精子计数,并统计精子畸形情况。结果与对照组比较,100μg/kgTBTCl染毒组小鼠的精子计数显著下降(P<0.01),各剂量TBTCl染毒组小鼠精子畸形率显著升高(P<0.05,P<0.01);且随着TBTCl染毒剂量的升高,小鼠精子计数呈下降趋势,精子畸形率呈升高趋势。而各剂量TBTCl染毒组小鼠包皮分离时间仅略有延迟。结论围产期低剂量TBTCl暴露可导致青春期雄性小鼠精子质量下降。Objective To explore the effects of perinatal exposure to low-dose tributylin chloride(TBTC1) on the quality of sperm in the male offspring mice in adolescence. Methods A total of 20 SPF health adult KM mice were given TBTCI(0,1,10 and 100 μg/kg, respectively, five mice each group) by gavage from the 6th gestation day until the end of lactation(21 days of age); the control mice received an equal volume of corn oil as vehicle(1 ml/kg). The males were examined daily at the same time of day starting on postnatal day (PND) 28 for completion of preputial separation (PPS) and the age in day of PPS was recorded. On postnatal day (PND) 49, the epididymis was separated for the sperm court and calculating the rate of sperm malformation. Results Compared with the control group ,the sperm count decreased significantly in 100 μg/kg group ,the abnormal rates of sperm significantly increased in all TBTC1- treated groups(P〈0.05 ,P〈0.01, respectively). A decreasing trend in the sperm count and an increasing trend in the rate of sperm malformation were observed following the TBTC1 dose increasing. Though the age of PPS was slightly delayed, there was no significantly difference between the TBTCl-treated groups and the control. Conclusion Perinatal exposure to low-dose TBTC1 can decrease the quality of sperm in the male offspring mice in adolescence.
