摘 要:目的研究油酸(oleic acid,OA)及其反式异构体反油酸(elaidic acid,EA)对人成熟脂肪细胞脂联素mRNA表达的影响。方法原代培养人脂肪组织来源的间质干细胞(human adipose tissue-derived stromal cells,hATSCs),诱导分化为成熟脂肪细胞,分别暴露于0(对照)、1、5、25μmol/L的OA和EA溶液72 h。采用实时定量PCR(real time PCR)方法检测脂联素mRNA的相对表达水平。结果与对照组比较,当人成熟脂肪细胞暴露于5、25μmol/L OA时,脂联素mRNA表达水平降低,差异有统计学意义(P<0.05);当人成熟脂肪细胞暴露于1、5、25μmol/L EA时,脂联素mRNA表达水平升高,差异有统计学意义(P<0.05)。且人成熟脂肪细胞脂联素mRNA表达水平随着OA染毒浓度的升高而下降,随着EA染毒浓度的升高而上升。与相同暴露剂量的OA组比较,EA暴露组人成熟脂肪细胞脂联素mRNA表达水平均高,差异有统计学意义(P<0.05)。结论 OA具有抑制人成熟脂肪细胞脂联素mRNA表达的作用,而EA具有增强作用。Objective To observe the impact on the adiponectin mRNA expression of human adipocytes by oleic acid (OA) and elaidic acid(EA). Methods Human adipose tissue-derived stromal cells(hATSCs) were cultured primarily,then were induced to differentiate into adipoeytes, which were exposed to OA and EA with different concentration for 72 h, 0μmol/L as control group, 1, 5 and 25 IxmolfL as exposure groups. Real time-PCR was used for mRNA detection of adiponectin mRNA expression. Results Compared with the control group, the adiponectin mRNA expression levels were decreased in the condition that human adipocytes were exposed to 5 and 25 μmol/L of OA, the difference were significant (P〈0.05); while the levels were increased in the condition that human adipoeytes were exposed to 1, 5, 25 μmol/L of EA, the difference were significant (P〈0.05). And the levels declined with the increase of the OA exposure concentrations, and increased with the increase of the EA exposure concentrations. Furthermore, compared with the adiponectin mRNA expression levels exposed to the same concentration of OA and EA in different groups, EA groups were significantly higher than OA groups (P〈0.05). Conclusion The results showed evidence of distinct effects of OA and EA on the expression of adiponectin in human adipocytes; OA can inhibit adiponectin mRNA expression ,yet EA has an enhanced effect.
