Culture Conditions Optimization for Antimicrobial Substances Production by Antagonistic Strain P-13 against Bacterial Spot Diseases of Muskmelon

摘  要:从新疆甜瓜根际土壤中分离到1株甜瓜细菌性斑点病拮抗菌P-13,对其培养基组成和摇瓶发酵条件进行优化。采用Plackett-Burman设计和最陡爬坡试验设计筛选培养基主要影响因子及其最佳区域,然后利用旋转中心组合和响应面分析确定最优培养基组成。最优培养基组成为葡萄糖15g/L、可溶性淀粉5g/L、蛋白胨3g/L、(NH4)2SO43.206g/L、CaCO36g/L、K2HPO40.6g/L、MgSO40.6g/L、NaCl 0.158g/L、FeSO40.006g/L和ZnSO40.002g/L。最佳摇瓶发酵条件为培养基初始pH 7.0、发酵温度28℃、摇瓶装液量为每500mL摇瓶装液60mL、发酵时间5.0d和接种量为φ=4%。发酵上清液对细菌性角斑病菌(Pseud-omonas syringae pv.lachrymans)P4的抑菌圈直径为30.51mm。Antagonistic strain P-13,with antimicrobial activity against muskmelon bacterial spot pathogens,was isolated from the muskmelon rhizosphere soil samples in Xinjiang.The effect of medium components and fermental conditions in Erlenmeyer flask on production of antimicrobial substances by strain P-13 was investigated in our work.In the first optimization step,the Plackett-Burman design and the paths of steepest ascent were effective in searching for the major factors and the optimal region of medium components.In the second step,the concentrations of medium components were further optimized with central composite rotatable design and response surface analysis.The optimal concentrations were as follows: glucose 15 g/L,soluble starch 5 g/L,peptone 3 g/L,(NH4)2SO4 3.206 g/L,CaCO3 6 g/L,K2HPO4 0.6 g/L,MgSO4 0.6 g/L,NaCl 0.158 g/L,FeSO4 0.006 g/L and ZnSO4 0.002 g/L.In the third step,the best conditions for the strain P-13 in Erlenmeyer flask were initial pH 7.0,temperature 28 ℃,medium capacity 60 mL per 500 mL,inoculum size 4%(volume fraction) and growth period 5.0 days.The diameter of inhibition zone of fermental supernatant from strain P-13 against Pseudomonas syringae pv.lachrymans P4 was 30.51 mm.
