摘 要:目的建立野菊花的反相高效液相色谱(RP-HPLC)指纹图谱和品质评价方法。方法采用优选的样品前处理方法,选用Kromasil C18(5μm,4.6 mm×250 mm)色谱柱,以乙腈-0.05%磷酸溶液为流动相(梯度洗脱),体积流量为1.0 ml/min,检测波长为334 nm,建立野菊花的RP-HPLC指纹图谱;以花形为小花,且相似度大于0.95,作为优质药材的判定标准,对22批不同产地野菊花进行品质评价。结果建立了野菊花的RP-HPLC指纹图谱,获得包含18个共有指纹峰的共有模式图谱,结合经验鉴别和指纹图谱建立了新的质量控制模式。结论本文建立的野菊花RP-HPLC指纹图谱方法专属性强,精密度和重复性良好,为野菊花整体质量控制提供了有效手段。Objective To establish a method of fingerprint analysis and quality evaluation on Flos Chrysanthemi Indici. Methods An optimized method of sample preparation was used, RP- HPLC was adopted with Kromasil Cls(5μm,4.6 mm×250 mm) , acetonitrile-0.05% H3PO4 solution (gradient elution) was used as a mobile phase with a flow rate of 1.0 ml/min, and detection wavelength was set at 254 nm. Fingerprint of 22 batches of Flos Chrysanthemi Indici from different regions were determined and their quality was evaluated : the good-quality flowers were floret and similarity between them was greater than 0. 95. Results The fingerprint of Flos Chrysanthemi Indici by RP-HPLC was established, including 18 common peaks. Combined experiences identification with fin- gerprint to build new quality control mode. Conclusion With high specificity, precision and good repeatability, the established meth- od arovides an effective means for overall aualitv control of Flos Chrvsanthemi Indici.
