和肾络颗粒对5/6肾大部切除大鼠肾脏增殖细胞核抗原表达的影响

摘　　要：目的:观察和肾络颗粒治疗5/6肾大部切除慢性肾衰竭模型大鼠肾组织增殖细胞核抗原(PCNA)表达的影响,探讨其对肾纤维化的保护作用。方法:采用5/6肾切除模型大鼠,设立正常对照组、模型组、西药福辛普利组、和肾络低剂量组、中剂量组及高剂量组。检测血尿素氮(BUN)、血肌酐(SCr),取残肾组织行HE染色,光镜观察肾组织病理变化,免疫组化检测PCNA的变化。结果:中药组与福辛普利组BUN、SCr明显低于模型组;PCNA在术后表达较正常对照组升高,用药组表达明显低于模型组。结论:和肾络抑制PCNA在肾组织内的过度表达,延缓肾脏纤维化的进程。Objection ： To observe the effects of Heshenlou Granules on expression of proliferating cell nuclear antigen （PCNA） in 5/6 subtotal nephrectomy rats ,which can help to understand its mechanism of protective effect on renal fibro- sis. Methods：Firstly, we constructed 5/6 subtotal nephrectomy model, then divided models into the normal control group, the operation model group, Fosinopril group, Heshenluo low - dose, middle - dose and high - dose group. Blood urea ni- trogen（BUN）and Serum creatinine（ Scr）were tested. Take the remnant kidney tissue for HE staining. Light microscopy was used to detect renal pathological changes. The expression of PCNA in renal tissue was tested by immunohistochemis- try. Results：Compared with the untreated 5/6 nephrectomized group, BUN and SCr of Heshenluo groups and Fosinopril group were substantially decreased（ P 〈 0.05 ）. The expression of proliferating cell nuclear antigen（PCNA） in model group was higher than that in normal group. It was less in Heshenluo groups and Fuxipuli group. Conclusion： Heshenluo can downregulate the expression of PCNA in kidney,which may slow down the process of renal fibrosis.
