摘  要:目的:观察糖痹康大鼠含药血清对高糖造成人脐静脉内皮细胞(HUVEC)损伤的保护作用。方法:以CCK-8法观察不同浓度葡萄糖对人脐静脉内皮细胞活力的影响;根据实验结果选择葡萄糖浓度为5.55mmol/L组为正常组,葡萄糖浓度为25mmol/L组做为模型组。然后制备正常组,弥可保组,糖痹康组的大鼠含药血清,将含药血清作用于高糖损伤的人脐静脉内皮细胞,分为正常组、高糖模型组、高糖弥可保组、高糖糖痹康低、中、高剂量组,并通过测定培养液中丙二醛(MDA)、乳酸脱氢酶(LDH)、一氧化氮(NO)含量与超氧化物歧化酶(SOD)活力,探讨糖痹康对血管内皮细胞损伤的保护作用。结果:与高糖模型组比较,糖痹康含药血清能减少细胞MDA、LDH释放量,提高SOD活力及NO含量。结论:糖痹康对高糖损伤的血管内皮细胞有一定的保护作用,间接的起到对周围神经的保护作用。Objective : To investigate the protective effects of Tangbikang Formula on human umbilical vein endothelial cells( HUVEC )injured by high - glucose. Methods : CCK - 8 was used to observe the viability of human umbilical vein endothelial cell in difierent glucose concentrations; according to the results, the glucose concentration of 5.55mmol/L group was selected as nornml group,the glucose concentration of 25retool/L group was as a model group. Then Normal group,Methycobal group and Tangbikang group were prepared. The diffenent serum which contained drags was given to the high- glucose- damaged human umbilical vein endothelial cells, and these cells were divided into normal group, model group, Methycobal group,Tangbikang low dose group,Tangbikang medium dose group,Tangbikang high dose group. By measuring the eontents of malundialdehyde( MDA ), lactate dehydrogenase( LDH ) ,nitric oxide(NO) and the activity of superoxide dismutase enzyme(SOD) in cell cuhure st,pernatant, the protective effect and mechanism of serum with Tangbi- kang Formula on injured HUVEC was assessed. Result: The serum with Tangbikang Formnula decreased the levels of MDA anti LDH and enhanced the activity of SOD and the level of NO in HUVEC injured by high -glucose. Conclusion :Tang- bikang Formula could prevent vascular endothelial cells from injury induced by high -- gluc.se,indirectly playing a role in prolection of peripheral neuropathy.
