摘  要:目的:观察止咳平喘合剂对哮喘大鼠气道炎症及血清白介素12(Interleukin-12,IL-12)、白介素13(Interleukin-13,IL-13)水平的影响,探讨其抑制哮喘气道炎症的作用机制。方法:用改良的卵白蛋白多点注射致敏与雾化吸入激发法建立大鼠哮喘模型。将40只雄性SD大鼠随机分为4组,每组10只,分别为空白对照组、哮喘模型组、强的松对照组和中药治疗组(止咳平喘合剂组)。实验第15天起,空白对照组、哮喘模型组分别给予生理盐水10mL.kg-1,强的松对照组给予强的松混悬液10mg.kg-1,中药治疗组给予止咳平喘合剂15g.kg-1,每天1次给药,连续5天。末次雾化激发后24h处死各组大鼠留取标本。镜下观察各组哮喘大鼠肺组织病理形态变化及支气管肺泡灌洗液(BALF)中白细胞、嗜酸粒细胞(EOS)数量;用双抗体夹心酶联免疫吸附试验(ELISA)法测定大鼠血清中IL-12、IL-13含量。结果:与哮喘模型组相比,中药治疗组大鼠BALF中白细胞总数和EOS数量均显著降低,差异有统计学意义(P<0.01);与哮喘模型组相比,中药治疗组大鼠肺组织气道黏膜及黏膜下层的炎性细胞浸润明显减轻;中药治疗组大鼠血清中细胞因子IL-12浓度明显高于哮喘模型组(P<0.01),而IL-13浓度明显低于哮喘模型组(P<0.01)。结论:止咳平喘合剂可通过提高细胞因子IL-12水平、降低IL-13的水平,来调节细胞因子失衡,从而减轻哮喘大鼠的气道炎症。Objectives: To observe the effects of Zhike Pingchuan Mixture on airway inflammation and levels of IL- 13 and IL - 12 and discuss the mechanism of that inhibition of airway inflammation in asthma. Methods : We used modified ovalbumin sensitized with multi - point injection atomization inhalation excitation method to establish the rat model of asthma. Fourty male SD rats were randomly divided into four experimental groups (10 in each) : normal control group, asthma model group, positive control group ( prednisone group) and Chinese medicine treatment group ( Zhike Pingchuan Mixture group). From the 15th day, the normal control group and the asthma model group were received normal saline with 10mL ~ kg- 1. The positive control group was given prednisone in 10mg ~ kg-1 while Chinese medicine treatment group was received 7.5g ~ kg-1 of Zhike Pingchuan Mixtureonce a day( each group was given once a day for 5 days). We killed rats and took samples of rats in different groups after the last 24 hours atomization excitation. We intesvigated the change of eosinophils count,the total count of leukocyte in BALF and the change of lung tissue of each group by micro- scope. The levels of IL - 13 and IL - 12 in blood serum were measured by ELISA. Results : Compared with asthma model group, the count of EOS and the total count of leukocyte in BALF of the Chinese medicine treatment group were signifi- cantly reduced(P 〈0.01 ). Compared with the asthma model group,airway mucosa and submucosa infiltration of inflam- matory cells in lung tissue of rat in Chinese medicine treatment group was significantly reduced . Compared with asthma model group,the concentration of cytokines IL - 12 in rat serum was significantly higher than that in Chinese medicine treatment group ( P 〈 0.01 ), while IL - 13 was significantly lower ( P 〈 O. 01 ). Conclusions : Zhike Pingchuan Mixture can increase level of cytokine IL- 12, decrease IL- 13 level, regulate cytokine imbalance, so as to reduce the airway infl
