摘  要:目的:探讨黄芪水煎剂(AE)对肾缺血再灌注损伤大鼠肾小管细胞凋亡的影响及机制。方法:雄性SD大鼠随机分为假手术组(Sham)、缺血再灌注损伤组(IR)及IR+不同剂量(0.5、1.0、2.0g/kg)AE预处理组(每组8只),夹闭双侧肾动脉45min再灌注6h后采用原位末端标记法(TUNEL)检测肾小管细胞凋亡,荧光试剂盒检测肾组织Caspase 3活性;实时定量PCR法检测Bcl-2、Bax mRAN表达。结果:与Sham组相比,IR组凋亡细胞数、Caspase 3活性及Bax mRNA表达均显著升高,而Bcl-2 mRNA表达明显降低(P<0.05);AE预处理组凋亡细胞数、Caspase 3活性及Bax mRNA表达均明显减少,而Bcl-2mRAN表达明显升高(P<0.05)。结论:AE能显著抑制IR大鼠肾小管细胞凋亡,机制可能与其调节Bcl-2、Bax表达有关。Objective:To study the effects of Astragalus extract(AE)on apoptosis of tubular epithelial cells in a rat model of renal ischemia/reperfusion(IR)injury and explore its possible mechanisms.Methods:Male Sprague-Dawley rats were randomized into sham operation group,IR group and IR + AE(0.5,1.0,2.0g/kg)treatment group.After clamping bilateral renal pedicles for 45 minutes and allowing reperfusion for 6 hours,the apoptosis of tubular epithelial cells was assessed by terminal deoxynucleotidyl transferase-mediated deoxy-UTP nick end labeling(TUNEL)method.Caspase 3 activity was detected by the fluorescent detection system.The mRNA expressions of Bcl-2 and Bax were examined by real-time PCR.Results:Compared with sham group,IR group had significant increase on apoptosis of tubular epithelial cells,Caspase 3 activity and Bax mRNA expression as well as decrease on Bcl-2 mRNA expresion,which was improved by AE pretreatment in a dose-dependent manner.Conclusion:AE inhibited apoptosis of tubular epithelial cells in a rat model of IR injury,which was possibly involved in regulation of Bax/Bcl-2 expression.
