DBP及E4BP4对拓扑异构酶Ⅰ的转录调节作用

摘　　要：目的探讨DBP(D site of albumin promoter(albumin D-box)binding protein)及E4BP4(腺病毒E4启动子结合蛋白-4)是否参与拓扑异构酶Ⅰ(TOPOⅠ)的调节作用。方法运用荧光素酶转录活性测定DBP和E4BP4对mTopoⅠ的转录活性,凝胶阻滞分析检测DBP及E4BP4蛋白和TopoⅠ核酸相互作用。结果荧光素酶转录活性测定实验显示DBP及E4BP4均能促进TopoⅠ基因的转录活性;凝胶阻滞分析实验直接证实DBP及E4BP4与TOPOⅠ启动子区域D-box活性区域结合发挥活性作用。结论 DBP及E4BP4参与调节拓扑异构酶Ⅰ的表达。Objective To explore the influence of D site of albumin promoter（albumin D-box） binding protein（DBP） and E4 promoter binding-protein 4（E4BP4） on transcription regulation of topoisomerase Ⅰ.Methods The effects of DBP and E4BP4 on mTopoⅠ expression were tested by cell transfection and luciferase assay.DNA（mTopoⅠ）–protein（DBP,E4BP4） binding was examined by gel-shift analysis.Results Luciferase assays indicated that mTopoⅠ gene expression was directly promoted by DBP/E4BP4.Gel-shift assay showed that D-site was a direct target of DBP and E4BP4.Conclusion DBP and E4BP4 are involved in the regulation of expression of topoisomerase Ⅰ.
