摘  要:目的:探讨Caspase-3在维A酸(RA)致小鼠胚胎神经管畸形发生过程中的表达及意义。方法:40只未经产昆明种小鼠随机分为实验组和对照组各4小组,每小组5只。分别在花生油及RA处理18、42、66、90 h后剖腹取胎、剥离头部神经管,Western-bloting法检测Caspase-3的表达变化。结果:Caspase-3在正常胚胎神经管组织中的表达存在时间依赖性。对照组Caspase-3在花生油灌服后18 h表达升高,42 h和66 h时表达最强,之后表达逐渐降低。实验组Caspase-3在RA处理后42 h表达水平显著低于对照组(P<0.01),66 h和90 h时表达水平均显著高于对照组(P<0.01)。结论:RA改变了昆明种小鼠胚胎神经管中Caspase-3的正常时间表达模式,这种异常的基因表达模式可能参与了RA诱导的神经管畸形发生。Objective:To explore the expression and significance of Caspase-3 in mouse with embryonic neural tube defects (NTD) induced by retinoic acid (RA). Methods:The 40 mice were randomly divided into test and control groups. The neural tubal samples were harvested at 18,42,66 and 90 h after RA or arachis oil administration. Western blotting was used to detect the expression of Caspase-3. Results :The expression of Caspase-3 displayed time dependence in the normal fetal neural tube tissue. In control group, the expression level of Caspase-3 increased at 18 h, and achieved the most peak at 42 and 66 h, then decreased slowly after arachis oil administration. In test group, the expression levels of Caspase-3 were significant lower than that of control group at 42 h ( P 〈 0.01 ) , and significant higher than that of normal group at 66 h and 90 h after treatment with RA ( P 〈 0.01 ). Conclusions : RA can change the expression pattern of Caspase-3 in embryonic neural tube of mouse, The Caspase-3 alteration may participate in the formation of NTD induced by RA.
