白细胞介素-6转化生长因子-β_1在2型糖尿病大鼠肺组织中表达变化的研究

摘　　要：目的通过检测白细胞介素(IL)-6、转化生长因子(TGF)-β1在2型糖尿病大鼠肺组织中表达变化,探讨其在糖尿病肺部炎症损伤中的作用。方法通过高糖、高脂饮食加腹腔注射小剂量链脲佐菌素(STZ)的方法,建立2型糖尿病大鼠模型,采用光镜动态观察12周、24周时对照组及糖尿病组的大鼠肺组织的形态学改变,用Masson三色染色观察肺组织胶原沉积情况,应用免疫组织化学方法检测各组大鼠肺组织IL-6、TGF-β1动态表达变化情况。结果糖尿病组大鼠肺组织结构紊乱,肺泡间隙增厚,肺间质和血管周围有炎症细胞浸润,胶原含量明显增多,糖尿病组大鼠肺组织IL-6、TGF-β1表达增多。结论糖尿病大鼠肺组织出现了炎症改变,糖尿病大鼠肺炎症的发生可能与IL-6、TGF-β1的含量增加有关。Objective To study the changes of IL-6 and TGF-β1 in lung tissue of diabetic rats and to investigate the role in pneumonia of diabetic rats. Method The experimental type 2 diabetic rats were yielded by injecting streptozotocin （STZ） and feeding with high fat and glucose food, then the alteration of morphology in the lung of rats were observed by the light microscope in the control group, the DM group during 12w and 24w, respectively, the collagen accumulation of lung by masson trichrome staining and alteration of IL-6,TGF-β1 in lung by immunohistochemistry. Results The organism structure of lung in the DM rats distributed deranged, bronchus wall and alveolar wall were thicken, alveolar epithelial cells were indistinct, desmocytes companied with inflammatory cell infiltration. The amount of collagen fiber increased. The expression of IL-6 and TGF-β1 increased in lung tissue of diabetic rats. Conclusion There were pulmonary inflammation in diabetic rats. The increasing level of IL-6, TGF-β1 in lung tissue of diabetic rats might be related to pulmonary interstitial inflammation of diabetic rats.
