摘  要:目的研究妊娠晚期母鼠脱水对成年子代大鼠脑肾素-血管紧张素系统(RAS)及其对水盐代谢调控的影响。方法妊娠母鼠随机分成对照组(Con)及脱水组(WD),脱水组于妊娠晚期脱水3 d,检测其成年子代鼠(5个月)血氧分压(PO2)、二氧化碳分压(PCO2)、pH值、血钠离子(Na+)和钾离子(K+)浓度及血浆渗透压(Osm)水平;检测其前脑血管紧张素受体(AT1R、AT2R)蛋白表达的水平,并记录成年子代鼠脑室给予血管紧张素Ⅱ(AngⅡ)后2 h内氯化钠(1.8%NaCl)和水的摄取量。结果妊娠晚期母鼠脱水3 d后,其成年子代鼠血PO2、PCO2、pH值、血Na+和K+浓度及Osm水平的差异均无统计学意义(均P>0.05);子代雌、雄大鼠前脑组织中AT1R蛋白表达水平均显著升高(均P<0.01),AT2R蛋白水平在子代雌鼠明显升高(P<0.05),雄鼠的差异无统计学意义(P>0.05);但雄鼠前脑的AT1/AT2蛋白表达比率明显升高(P<0.05),雌鼠的差异无统计学意义;子代雌、雄大鼠脑室给予AngⅡ后2 h内摄盐量的差异无统计学意义(P>0.05),子代雄鼠摄水量显著增多(P<0.01)。结论妊娠晚期母鼠脱水可引起其成年子代水盐代谢的改变,该变化可能与前脑血管紧张素受体表达上调有关。Objective To determine the effect of maternal water deprivation during late gestation on water-electrolyte metabolism related to renin-angiotensin system(RAS) in the offspring rats. Methods Maternal rats were deprivated of water for 3 days during late gestation period, The arterial blood PO2, PCO2 ,pH ,Na+/K + concentrations andplasma osmotic pressure of 5-month old offspring were measured. Angiotensin receptor proteins expression in the forebrain were determined by western blot, And intake of 1.8% NaC1 and water in the following 2 hours after intracerebroventricular (i. c. v) angiotensin lI were measured. Results Blood values ( PO2 , PCO2, pH, Na ~/K ~ concentrations and osmolality) were not changed by maternal water deprivation during late gestation. AT1 R protein expression in the forebrain was increased significantly in both male and female adult offspring, while AT2R protein expression was in- creased only in females. Their ratio of AT1/AT2was increased in males, but not females. I. c. v angioten- sin I1 -induced water intake in male increased significantly, but not female. Conclusions Maternal water deprivation during late gestation could alter water-electrolyte metabolism, which was associated with thechanges of the expression of AT1 and AT2 receptors in the forebrain of the adult offspring.
