探讨EOS染色剂对血迹DNA检验的影响

摘　　要：目的探讨被EOS染色剂处理后的血迹进行DNA检验的初步方法。方法制备EOS染色剂处理的血迹样本,分别采取纯水擦拭、75%酒精擦拭,或手术刀刮取血痕浸泡于纯水中、75%酒精中,然后提取DNA进行下一步检测。结果采用刀刮取血痕浸泡于75%酒精中,提取的DNA检测结果较好。结论初步实验显示,经EOS染色剂处理过的血迹,可参考刀刮取血迹置于75%酒精浸泡后提取DNA做下一步检测。Objective To investigate the effect of fingerprint reagent EOS to DNA test and explore methods for testing DNA from blood fingerprints treated with EOS. Methods The blood samples treated by EOS were wiped with water, wiped with alcohol, and scraped with scalpel, then the DAN were analyzed with ABI 3100 Genetic Analyzer and the results were compared. Results STR loci were genotyped successfully in the sample which was scraped and alcohol treated. Conclusion Scraping the samples and then treating with alcohol is the best way to extract DNA from the blood fingerprint treated by reagent EOS.
