摘 要:目的探讨蕨麻正丁醇提取物对心肌缺血/再灌注(I/R)损伤大鼠肿瘤坏死因子-α(TNF-α)的影响。方法50只Wistar大鼠按随机数字表法分为假手术组、模型组和蕨麻提取物高、中、低浓度预处理组,每组10只。右心房取血,用酶联免疫吸附试验(ELISA)检测血清TNF-α含量,用逆转录-聚合酶链反应(RT-PCR)、蛋白质免疫印迹法(Westernblotting)检测心肌组织中TNF-α mRNA和蛋白表达。结果与假手术组比较,模型组及蕨麻提取物中、低浓度预处理组血清TNF-α水平及心肌组织TNF-α mRNA[吸光度(A)值]和蛋白表达均明显升高,以模型组升高更显著[血清TNF-α(pg/L):93.8±5.3比46.5±4.2,心肌组织TNF-α mRNA:0.79±0.11比0.37±0.10,心肌组织TNF-α蛋白:0.92±0.09比0.49±0.11,均P〈0.05];与模型组比较,蕨麻提取物高、中、低浓度预处理组TNF-α均明显降低,以蕨麻提取物高浓度预处理组降低更显著(血清TNF-α:44.0±2.9,心肌组织TNF-α mRNA:0.33±0.09,心肌组织TNF-α蛋白:0.45±0.18,均P〈0.05)。结论TNF-d在大鼠心肌I/R损伤过程中表达上调,用蕨麻正丁醇提取物进行干预可降低TNF-α的表达。Objective To investigate the effects of Potentilla anserine L. (Pc) extract on the level of tumor necrosis factor-α(TNF-α ) in rats with acute myocardial isehemia/reperfusion (I/R) injury. Methods Fifty Wistar rats were randomly divided into sham operation group, model group and Pa high-dose group, middle-dose group and low-dose group. Right atrial blood was collected to detect the serum TNF-α content by enzyme-linked immunosorbent assay (ELISA), and the expressions of TNF-α mRNA and protein of the tissues were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting. Results Compared with the sham operation group, the expressions of the absorbance (A) value of TNF-α mRNA in serum and myocardial tissues in model group, Pa high-dose group, middle-dose group and low-dose group were increased significantly, and the most significant one was the model group [ serum (μg/L) : 93.8±5.3 vs. 46.5±4.2, tissue mRNA : 0.79±0.11 vs. 0.37±0.10, tissue protein : 0.92±0.09 vs. 0.49±0.11, all P〈0.05]. Compared with the model group, TNF-α reduced significantly in Pa high-dose group, middle-dose group and low-dose group, and the most significant one was the high-dose group (serum : 44.0±2.9, tissue mRNA : 0.33±0.09, tissue protein: 0.45±0.18, all P〈 0.05). Conclusions TNF-ot is up-regulated in rats with acute myocardial I/R injury. Pa extract can be used to intervene and reduce the expression of TNF-α.
