摘  要:目的:通过对人参生长过程中5种酶活力进行测定,为人参的鉴定和优选提供理论依据。方法:分别于展叶期、开花期、绿果期、红果期、枯萎期采取人参根样品。采用中性缓冲液提取总蛋白,应用分光光度法对人参各个生长时期中硝酸还原酶(nitrae reductase,NR),超氧化物歧化酶(superoxide dismutase,SOD),谷丙转氨酶(glutamic-pyruvic transaminase,GPT),葡萄糖磷酸异构酶(glucose pHospHate isomerase,GPI)和延胡索酸酶(fumarase)5种酶的活力进行测定。结果:在人参根生长发育过程中,NR在展叶期活力最高;GPT和延胡索酸酶分别在展叶期、结果期、果熟期出现活力峰值;GPI在结果期和果熟期活力均较高。结论:NR,SOD,GPT,GPI,延胡索酸酶的活力的测定可以为人参酶的进一步研究奠定基础。Objective: To provide the basis for identification and selection of ginseng radix through determining the 5 kinds of enzymatic activities at different growth stages.Methods: Ginseng samples were respectively collected at stages of the exhibition leaf,flowering time,green fruiting,red fruits period,and wither period.Neutral buffer solution was used to extract the enzyme solution of Radix Ginseng.Spectrophotometric method was used to determine activities of ginseng nitrae reductase(NR),superoxide dismutase(SOD),glutamic-pyruvic transaminase(GPT),glucose phosphate isomerase enzyme(GPI) and fumaric acid enzyme(fumarase) at different stages.Results: NR activity was the highest in the spreading stage.GPT and Fumarase activities had two peaks at the spreading,fruit setting and ripening stages.GPI activity was high at the fruit setting and ripening stages.Conclusion: The enzymatic activities of NR,SOD and GPT,GPI,and fumarase can be used for the determination of ginseng enzymes in further researches.
