HPLC法测定瘤果黑种草子中常春藤皂苷元及齐墩果酸含量

摘　　要：目的建立HPLC法同时测定瘤果黑种草子中常春藤皂苷元及齐墩果酸的含量。方法色谱柱:HypersilODS C18(150 mm×4.6 mm,5μm);流动相:乙腈-水-冰醋酸-三乙胺(85∶15∶0.04∶0.02);流速:0.5 ml/min;检测波长210 nm。结果 TLC能检出齐墩果酸、常春藤皂苷元相应的斑点;HPLC法测定常春藤皂苷元、齐墩果酸分别在22.76～227.6、40.28～201.4μg/ml范围内线性关系良好,r值分别为0.9998、0.9997,平均回收率分别为94.6%、93.8%,RSD分别为1.8%、1.2%。结论本法可用于瘤果黑种草子中皂苷的质量控制,方法简单,结果准确,灵敏度高。Objective To establish an HPLC method for determination of the content of hederagenin and oleanic acid in seeds of Nigella glandulifera.Methods HPLC analysis was carried out using Hypersil C18（150 mm×4.6 mm,5 μm） column and acetonitrile-water-glacial acetic acid-triethylamine（85∶ 15∶ 0.04∶ 0.02） as the mobile phase.The flow rate was 0.5 ml/min and the detection wavelength was 210 nm.Results The linear range of hederagenin and oleanic acid was respectively 22.76-227.6 μg/ml（r=0.9998）and 40.28-201.4 μg/ml（r=0.9997）.Their average recovery was respectively 94.6% and 93.8%,and RSD was 1.8%,1.2%.Conclusion This method is simple,quick and accurate,which can be used for quality control of saponins in seeds of Nigella glandulifera.
