摘  要:目的:使用外源性标记法研究氯高铁血红素在大鼠体内的铁吸收。方法:以58Fe标记的氯高铁血红素分别以40mg/kg和2mg/kg的剂量灌胃和静脉注射给予大鼠,并在其给药前和给药后系列时间点采样,用电感耦合等离子体质谱仪(ICP-MS)法测定各个时间点58Fe的血药浓度,测得的血药浓度通过DAS2.1.1求算药动学参数。用58Fe的绝对生物利用度评价氯高铁血红素在大鼠体内的铁吸收。结果:源于氯高铁血红素中的58Fe在灌胃给药后1.5h吸收达峰,峰浓度为(220±89)μg/L,在此剂量下源于氯高铁血红素中的58Fe绝对生物利用度为0.93%。结论:外源性标记法可以用于评价氯高铁血红素在大鼠体内的铁吸收,源于氯高铁血红素的58 Fe绝对生物利用度为0.93%。AIM. To evaluate the pharmacokinetic characteristics and bioavailability of 58 iron originating from hemin in rats. METHODS: A rapid, sensitive and specific method with inductively coupled plasma mass spectrometry (ICP- MS) has been developed for the determination of s8 iron in bio-samples. The study was performed in male rats at a single dose of 40 mg/kg and 2 mg/kg separately by oral administration and in- travenous injection. Pharmacokinetic parameters were calculated by non-compartmental method using DAS 2.1.1 program and the dose of ^58 iron was used in the calculation of these parameters. RESULTS: Results showed that $8 iron origina-ting from hemin was poorly absorbed and the absolutely bioavailability was 0.93%. Plasma concentrations of ^58iron reached the peak at 1.5 h following oral administration to rats and the peak concentration was (220 ± 89) μg/L. CON- CLUSION: This research successfully investigated the pharmacokinetic characteristics of $8 iron originating from heroin using extrinsically label method and the absolutely bioavailability was evaluated.
