摘 要:目的探讨Ang-1和Ang-2及其受体Tie-2在鼻咽癌(nasopharyngeal carcinoma,NPC)患者血循环中的表达及其临床意义。方法应用酶联免疫吸附试验检测2009年5月至2010年6月本科收治的低分化鳞状细胞癌患者68例,其中男性52例,女性16例,中位年龄42(19~65)岁(初治32例,完全缓解20例,复发16例)及20例健康对照者血清Ang-1和Ang-2及其受体Tie-2水平。结果初治组和复发组NPC患者血清Ang-1和Ang-2及其受体Tie-2的水平均高于完全缓解组及健康对照组(P<0.01),而这些分子的含量在完全缓解组和健康对照组比较无明显差异(P>0.05)。Ang-2及Tie-2的水平在NPC患者不同T分期、临床分期比较有差异(P<0.05),并随着NPC进展呈上升趋势;Ang-1水平在不同T分期、临床分期比较差异不显著(P>0.05);Ang-2水平在不同N分期比较有差异(P<0.05)。转移NPC患者血清Ang-2及其受体Tie-2水平明显高于无转移组(P<0.01)。Ⅰ+Ⅱ期NPC患者血清Ang-1/Ang-2比值高于Ⅲ+IVa期(P<0.01)。结论 Ang-1和Ang-2及其受体Tie-2在鼻咽癌的发生、发展过程中起重要作用,并与肿瘤的侵袭转移有密切关系。Objective To investigate the serum levels of angiopoietin-1 (Ang-1), angiopoietin-2 (Ang-2) and their receptor, tyrosine kinase with immunoglobulin-like and epidermal growth factor homology domains Tie-2 in patients with nasopharyngeal carcinoma ( NPC ). Methods A total of 68 patients ( 32 untreated cases, 20 cases with complete remission and 16 recurrent cases) with pathologically identified NPC (all poor differentiated squamous cell carcinoma) who were hospitalized in our department from May 2009 to June 2010 were enrolled in this study. Their serum samples and samples from 20 healthy age- and sex-matched volunteers (control group) were collected. The serum levels of Ang-1, Ang-2 and Tie-2 were measured by enzyme-linked immunosorbent assay (ELISA). Results The serum levels of Ang-1, Ang-2 and Tie-2 in untreated and relapsed NPC groups were significantly increased than those in the complete remission group and normal control (P 〈 0.01 ). But no obvious difference was found in the 2 latter groups (P 〉 0.05 ). Only the levels of Ang-2 and Tie-2 were closely correlated with T and clinical stages ( P 〈 0.05 ). The serum levels of Ang-2 and receptor Tie-2 tended to increase with the development of tumor. The serum level of Ang-2 was closely correlated with N stages (P 〈 0.05 ), but not with T and clinical stages ( P 〉 0.05 ). Serum levels of Ang-2 and receptor Tie-2 were significantly higher in tumor metastasis group than non-metastasis group (P 〈 0. 01 ). Moreover, the ratio of Ang-1 to Ang-2 was higher in NPC patients at I + II stages than those at m + IVa stages (P 〈 0.01 ). Conclusion Ang-1, Ang-2 and receptor Tie-2 play important roles in the progress of NPC, and may be involved in the invasion and metastasis of the tumor.
