水稻成熟种子组织培养体系的优化及建立

摘　　要：通过对3个粳稻品种成熟种子的表面消毒、愈伤组织诱导、分化及生根条件等对水稻成熟种子培养再生成株,对种子表面消毒剂、2,4-D浓度、继代培养时间等条件进行了优化。结果表明,在优化条件下,粳稻的绿苗分化率在90%以上,由此建立了一套高效、可靠、重复性好的水稻组织培养再生系统,为水稻遗传转化的顺利进行奠定了基础。Factors affecting the regeneration of rice plants from maturate seeds, such as sterilizing methods, induction,differentiation and rooting conditions, etc., were investigated.Varieties of sterilizing antiseptics, concentration of 2,4-D, time of secondary culture.The results showed that the optimal conditions for the frequency of green shootdifferentiation of the three breed were all more than 90%.Therefore a high efficient regeneration system from in vitro culture of maturate seed was established for genetic transformation of rice.
