摘  要:目的 观察多次腹腔注射丙泊酚(propofol,P)对小鼠自主活动和学习记忆能力的影响.方法 昆明种小鼠90只,分为镇静和记忆两实验大组,每实验大组再分为3组(n=15):NS组腹腔注射生理盐水,另两组分别腹腔注射丙泊酚10 mg/kg(P10组)、15 mg/kg(P15组).用自主活动仪检测每组小鼠第1、2、3天第2次给药15 min后5 min内的自主活动次数,用跳台仪观察小鼠最后1次给药后24、48、72 h的跳台潜伏期(step-down latency)和错误次数(error times).结果 镇静实验结果显示,在第1、2、3天,P10组小鼠自主活动次数与NS组比较差异无统计学意义(P〉0.05),而P15组活动减少(P〈0.01或P〈0.05);每组第1、2、3天之间差异没有统计学意义(P〉0.05).记忆实验结果显示:在24 h以及48 h,P10组、P15组与NS组相比跳台潜伏期缩短(P〈0.01或P〈0.05)、错误次数增多(P〈 0.05);72 h时,3组潜伏期和错误次数的差异均无统计学意义(P〉0.05).结论 小鼠对丙泊酚产生的遗忘作用敏感性高于其产生的镇静作用;多次应用丙泊酚会产生遗忘作用,但不会持续到48 h.Objective To observe the impacts of propofol (P) by intraperitoneal injection (ip) repeatedly on autonomic activities and learning and memory in mice. Methods 90 Kunming mice were randomly divided into 2 great test groups: sedative test group and memory test group. Each great test group were divided futher into 3 small groups: NS group (ip NS), P10 group (ip propofol 10 mg· kg^-1) , P15 group (ip propofol 15 mg · kg^-1), The autonomic activities apparatus was used to observe the autonomic activities in 5 thin of each group after 15 rain by ip medicine at the I st, 2nd and 3rd day; the step - down apparatus was used to observe the step - down latency and error times at 24 h, 48 h, 72 h after the last ip medicine. Results In sedative test, at the 1st, 2nd and 3rd day, compared with NS group, the autonomic activities of PJ0 group had no statistically significant difference ( P 〉 0.05 ) , but that of P15 group decreased ( P 〈 0.01 or P 〈 0.05 ) ; there were no statistical significances at the 1 st, 2nd or 3rd day with each group (P 〉 0.05). In the memory test, compared with NS group, at 24 h or 48 h, step - down latency shortened (P 〈 0.01 or P 〈 0.05 ) and the error times decreased (P 〈 0.05 ) of P10 group or P15 group, but that of the three groups had no statistically significant difference at 72 h ( P 〉 0.05 ). Conclusion Mice are more sensitive to the forgotten effects than the sedation of propo- fol; repeated applications of propofol can produce forgotten effects but can't last 48 hours.
