新鲜与陈旧外周抗凝血中miRNA表达变化的研究

摘  要:目的比较新鲜与陈旧外周抗凝血中miRNA(microRNA)表达的变化。方法取5份新鲜及6份陈旧外周抗凝血液样本,提取全血总RNA并测定其纯度与浓度,应用实时定量RT-PCR技术分析外周血中miRNA的表达变化。结果经过核酸蛋白定量仪检测,新鲜与陈旧外周抗凝血液中总RNA A260/280值为1.8~2.0范围内。实时荧光定量RT-PCR的结果显示:新鲜与陈旧的外周抗凝血液中miRNA RNAU6-2的相对拷贝数差异无统计学意义。结论新鲜与陈旧外周抗凝血液中miRNA RNAU6-2均有表达且无差异。Objective To compare the expression of miRNA(microRNA) between fresh and old peripheral anticoagulant blood.Methods 5 pieces of fresh blood samples and 6 pieces of old blood samples were chose respectively,extracted total RNA from the whole blood and measure its purity and concentration,and analyzed the microRNA expression using real-time quantitative reverse transcription polymerase chain reaction(real-time RT-PCR).Results With Micro-spectrophotometer test,A260/280 of total RNA was 1.8~2.0.The comparative number of copies of miRNA RNU6B-2 has no statistical difference between fresh and old peripheral anticoagulant blood.Conclusion There was no difference between miRNA RNU6B-2 expressed in fresh and old peripheral anticoagulant blood samples.
