胰岛素对大鼠急性全脑缺血再灌注损伤NSE及行为学变化的影响

摘　　要：目的探讨不同疗程胰岛素对大鼠急性全脑缺血再灌注损伤的影响及机制。方法将72只雄性Wistar大鼠随机分为3组:假手术组、对照组与胰岛素治疗组,按疗程每组分别分为1d组、3d组与7d组。采用四血管阻断法制备大鼠全脑缺血再灌注损伤模型。治疗组腹腔注射胰岛素2 IU/(kg.d)和50%葡萄糖注射液2g/(kg.d),对照组腹腔注射生理盐水2mL/(kg.d),治疗过程中监测血糖,每组动物治疗结束前检测行为学变化,结束时取血测定血清神经元特异性烯醇化酶(NSE)含量,并观察海马病理学改变。结果与对照组比较,治疗组大鼠学习记忆力明显提高,血清NSE浓度降低,脑组织缺血性损伤减轻。结论胰岛素可减轻大鼠全脑缺血再灌注损伤后脑组织损伤程度,且保护作用随疗程增加逐渐增强。Objective To observe effects and mechanisms of different course of treatment in insulin on cerebral ischemia reperfusion injury. Methods 72 male Wistar rats were randomly divided into 3 groups： shamoperated group, control group, and treated group. According to different course every group was divided into 3 groups： ld,3d,7d group. Four-vessel occlusion was used to establish global cerebral ischemia reperfusion model in groups. The treated group were intraperineally injected with biossynthetic human insulin 2 IU/（kg ~ d） and 50% glucose 2g/（kg ~ d） for aforementioned days. The control group rats were given with saline 2mL/（kg ~ d） in abdominal cavity. The blood glucose was monitored during treated days. The changes of action of rats were measured before killed. The concentration of neuron specific enolase（NSE） in blood serum was dectected. The hippocampus was separated for observation of pathology changes. Results Compared with the control group, the memory of rats in treated group was greatly improved, consentration of NSE in blood serum was significantly decreased, and the damage of cerebral was greatly alleviated. Conclusion Insulin can reduce the degree of damaged in brain of global cerebral ischemia reperfusion injury in rats.The protective effects of insulin increase gradually with the course of treatment.
