摘 要:目的探讨盐酸美金刚对兔受损视神经的保护作用。方法取健康大白兔66只,随机分为3组,对照组A组,损伤生理盐水组B组,损伤治疗组C组,B、C两组又随机分为损伤B1组、B2组(作病理切片用),损伤治疗组C1组、C2组(作病理切片用)。B、C两组兔双眼作视神经损伤模型后,C组给予盐酸美金刚药物治疗,剂量每天每只兔10mg,每天上午一次性给全部药物。B组给予生理盐水。分别于造模后1d、3d、7d、14d、21d、28d随机处死各组兔子,检测视网膜谷氨酸浓度变化及观察视网膜病理形态学变化。结果造模和用药后1d、3d、7d、14d损伤生理盐水组和损伤治疗组视网膜谷氨酸含量与对照组相比差异均有统计学意义(均为P<0.05),均较对照组高。损伤生理盐水组视神经损伤后谷氨酸浓度迅速增高,第3天达最高峰(13.90±0.55)mmol.L-1,与对照组(2.59±0.37)mmol.L-1比较差异有统计学意义(P<0.05),从第7天开始神经节细胞数量明显减少,细胞外形皱缩,核固缩。盐酸美金刚药物治疗组第3天谷氨酸含量(6.75±0.45)mmol.L-1明显低于损伤组(13.90±0.55)mmol.L-1,且差异有统计学意义(P<0.05)。伤后第1天、第7天和第14天时谷氨酸含量两组比较差异亦均有统计学意义(均为P<0.05)。结论盐酸美金刚药物能明显降低谷氨酸浓度从而减少细胞凋亡,保护受损视神经。Objective To investigate the protective effects of memantine hydro- chloride on rabbits injured optic nerve. Methods Sixty-six healthy rabbit were ran- domly divided into control group ( group A), injured saline group ( group B), injured treatment group ( group C) ,group B and C was randomly sub-divided into group BI and B2,group C1 and C2. After the dual eyes in group B, C established the optic nerve injury models, memantine hydrochloride drug therapy performed in group C, 10 mg of daily do- ses in each rabbit and daily morning disposable to all drugs,and saline was given to rab- bit in group B. The rabbits were sacrificed at 1 day,3 days,7 days,14 days,21 days and 28 days after modeling, the changes of retinal glutamate content and retinal pathologic morphous were detected. Results Compared with group A, there were statistical differences in retinal glutamate content in group B and C at 1 day,3 days,7 days and 14 days after modeling ( all P 〈 0.05 ). The glutamate concentration in group B increased rapidly after optic nerve injury,peaked at 3 days,which was ( 13.90 ±0.55) mmol L-I , compared with group A (2.59 ±0.37)mmol L-1 ,there was statistical difference (P 〈 0.05 ). From 7 days, number of ganglion cells significantly reduced with marcid cell shape and nucleus. The glutamate concentration at 3 days in group C was (5.75 ±0.45 ) mmol L-1 ,lower than that in group B,there was statistical difference(P 〈0. 05 ) ,and there were also statistical differences at 1 day,7 days and 14 days between two groups ( both P 〈 0.05 ). Conclusion Memantine hydrochloride drug can significantly reduce the glutamate concentration and decreased the apoptosis.
