摘  要:目的研究血压变异性(BPV)对Sprague Dawley(SD)大鼠主动脉及心肌组织瞬时受体电位通道6(TRPC6)表达的影响,探讨TRPC6与BPV的关系。方法 20只10周龄雄性SD大鼠随机分为两组,去窦弓神经(SAD)手术组和假手术组,每组10只。术后8周测量两组大鼠BPV、左心室质量指数(LVMI)以及主动脉和心肌组织中TRPC6的mRNA和蛋白表达。计算大鼠收缩压和舒张压的均数及标准差,并以标准差作为该时段的BPV。结果 SAD组24h收缩压标准差[(12.6±3.4)比(7.0±2.1)mmHg]及舒张压标准差[(8.2±3.8)比(3.4±0.5)mmHg]、LVMI[(2.19±0.06)比(1.71±0.04)mg/g]、主动脉及心肌组织中TRPC6的mRNA和蛋白表达均明显高于假手术组(均P<0.05)。结论 BPV增大时,大鼠主动脉及心肌组织TRPC6表达量增加。Objective To investigate the effect of blood pressure variability (BPV) on expression of transient recep tor potential channel 6(TRPC6) in aorta and myocardium in Sprague Dawley(SD) rats with sinoaortic denervation. Methods Twenty male SD rats were randomly divided into two groups underwent sinoaortic denervation ( SAD) and sham operation respectively at the age of ten weeks. Eight weeks after operation intervention, the BPV, left ventricular mass index (LVMI) and the expression of TRPC6 mRNA and protein in aorta and myocardium were detected. The mean and standard deviation of 24 h systolic and diastolic blood pressure were calculated, and standard deviation was served as BPV. Results Compared with the sham group, the 24 h systolic blood pressure standard deviation [(12.6±3.4)vs(7.0±2.1) mm Hg], diastolic blood pressure standard deviation [-(8.2± 3.8)vs(3.4q- 0.5) mm Hg], LVMI [-(2.19±0.06)vs(1.71±0. 04)mg/g, P〈0.05] and the expression of TRPC6 mRNA and protein in aorta and myocardium were significantly increased in SAD group (P 〈0.05, respectively ). Conclusion The BPV may associate with the increased expression of TRPC6 in aorta and myocardiun in SD rats.
