摘 要:目的了解慢性HBV感染者外周血单核细胞(PDMC)中CD4~+T细胞内IL-4和IFN-γ的表达情况,以测定Th0/Th1/Th2细胞的百分数,探明Th1、Th2细胞在慢性HBV感染中的作用。方法常规分离PBMC,在PMA、ionomycin、莫能菌素的刺激下,采用流式细胞检测(FACS)对慢性HBV感染者PBMC中CD4~+T细胞内IL-4和IFN-γ表达进行分析。结果正常对照组,2.3%-18.6%的CD4~+T细胞为Th1细胞,0.9%-9.2%为Th2细胞;在慢性HBV感染者外周血单个CD4~+T细胞中,以Th0细胞居多:而Th1细胞则随着慢性乙型肝炎肝脏炎症活动的加剧而明显增多,在炎症活动明显的慢性乙型肝炎患者中,Th1细胞百分数明显高于炎症活动呈静止状态的慢性乙型肝炎;Th2细胞在慢性HBV感染的不同阶段则较为恒定,但明显高于对照组。结论Th1细胞与炎症活动呈正相关,Th2细胞可能与HBV感染的慢性化相关联。Objective The ratio of Th0/Th1 /Th2 cells in chronic HBV-infected individuals was investigated by determining the expression of IL-4/IFN-γ in Single CD4^+ T cells isolated from peripheral blood. and the role of Th1 /Th2 cytokines in chronic HBV-infection was analyzed. Methods Peripheral blood mononuclear cells (PBMCs) from chronically HBV-infected individuals were isolated routinely, stimulated by PMA, ionomycin and monensin, and the production of IL-4/IFN-γby CD4^+ T cells in PBMC was determined by FACS analysis. Results The percentage of IFN-γ-producing T cells and IL-4 producing T cells ranged from 2.3% to 18.6% and from 0.9% to 9.2% respectively in CD4^+ cells in non-infected individuals The majority of CD4^+ T cells from PBMCs in chronic HBV-infected individuals were Th0 cells. The percentage of Th1 cells increased significantly with the hepatic inflammation activity The percentage of Th1 cells was higher in advanced stage than in stationary phase of chronic hepatitis B, The percentage of Th2 cells in CD4^+ T cells from HBV-infected individuals showed no significant difference at different stages, but was higher than that from controls. Conclusion The results indicated that Th1 phenotype cytokines were associated with hepatic inflammation activity of chronic hepatitis B and Th2 cells may be associated with the chronicity of HBV Infection.
