摘  要:目的分析慢性粒细胞白血病 ( CML)病人的克隆性增殖 T细胞及其 CDR3序列的特点。方法利用 RT- PCR扩增 3例 CML 外周血单个核细胞中 2 4个 TCR Vβ基因的 CDR3,了解 TCR VβT细胞的分布。阳性产物进一步经基因扫描分析 ,了解其克隆性。寡克隆的 PCR产物再进行序列分析。结果 3例病人仅存在 3~ 9个 TCR Vβ亚家族 T细胞 ,部分 Vβ亚家族T细胞呈寡克隆性增殖 ,Vβ2 1的寡克隆 T细胞见于全部的 3例病人中。结论 CML 病人外周血 TCR Vβ亚家族 T细胞出现倾斜性分布和克隆性增殖 ,这可能是 CML 中机体对恶性细胞所引起的特异性免疫反应 ,但对不同病人的 Vβ2 1寡克隆 T细胞序列分析并未能发现完全同源的ObjectiveTo investigate the clonal expansion and the feature of CDR3 sequence of T cells in patients with chronic myelogenous leukemia (CML)MethodsThe CDR3 of TCR Vβ 24 subfamily genes were amplified in peripheral blood mononuclear cells from 3 cases with CML using RTPCR, to observe the distribution of TCR Vβ subfamily T cells ,the PCR products were further analyzed by genescan technique to evaluate clonality of the detectable TCR Vβ T cells The products of the oligoclonal T cells were analyzed by sequencing to define the sequence of CDR3ResulstOnly 3~9 Vβ subfamily T cells could be identified in 3 CML cases Oligoclonal expansion T cells could be detected and Vβ21 oligoclonal T cells were identified in all 3 cases ConclusionsThe skew distribution and clonal expansion of TCR Vβ subfamily T cells could be found on the patients with CML It may be a feature of the host immune response for malignant cells in CML However, the completely homologous CDR3 sequence of Vβ21 oligoclonal T cells from different cases could not be detected [
