摘 要:目的:了解小剂量山茶籽联合雌二醇对去卵巢大鼠骨组织的影响,为中西医结合治疗I型骨质疏松症提供实验依据。方法:6月龄健康雌性大白鼠,分成假手术组(sham),去卵巢模型组(OVX),山茶籽组(Pr),雌二醇组(E2)和小剂量山茶籽醇提取物+雌二醇组(Ts+Ee)。取左股骨切片观察骨组织,取右股骨测量骨钙、磷和骨密度。结果:卵巢模型组的骨组织明显疏松,骨钙、磷低于Sham组,3个治疗组各时间的骨组织、骨钙、磷和骨密度与Sham组比较差异无统计学意义。小剂量的山茶籽联合雌二醇治疗能使去卵巢大鼠骨组织和骨钙、磷和骨密度恢复正常,与较大剂量的山茶籽组或较大剂量的雌二醇组相比无差异。结论:山茶籽对去卵巢大鼠的骨质疏松症的治疗效果良好;小剂量的雌二醇与山茶籽对去卵巢大鼠的骨质疏松症的治疗效果与单独使用较大剂量的山茶籽或较大剂量的雌二醇相比治疗效果相近。Objective: To explore the effect of low-dose extract of camellia seed combined with estradiol on the bone tissue in ovarieetomized rats, therefore, to provide experimental evidence for treating osteoporosis. Methods: 120 6-month-old healthy female rats were randomly divided into five groups including sham, ovariectomized (OVX), and three experimental groups according different treatments such as extract of camellia seed (5 g/g, intragastric administration), estradiol (E2) (200/μg/kg, 2 times per week, subcutaneous injection) and extract of camellia seed (2.5 g/g, intragastric administration)-E2 (100 μg/kg, 2 times per week, subcutaneous injection). All three treatments were performed 1 week after the ovariectomy. ()n the postoperative week 4, 8, 12 and 20, the left femurs of animals were dissected to detect contents of bone calcium and bone phosphorus, as well as bone density. Results: Compared to the sham, the bone density, contents of calcium and phosphorus in the femurs of OVX rats was significantly decreased. And, three different treatments led to recovery in bone density, contents of calcium and phosphorus, with no significant difference compared to those in sham. Moreover, statistical difference was undetectable be tween the animals treated with low dose extract of camellia seed and estradiol and those treated with high dose extract of camellia seed only or high-dose estradiol only. Conclusion: These findings suggest that extract of camellia seed is effective to treat the osteoporosis of rats. And, low-dose extract of camellia seed combined with estradiol has a similar pharmacological effect compared to high-dose extract of camellia seed only or estradiol only.
