摘 要:目的:研究日本血吸虫尾蚴感染鼠CD8α-CD11c+型树突状细胞(DC)在抑制过敏性哮喘中的作用。方法:用磁珠分选方法纯化日本血吸虫尾蚴感染鼠及正常鼠DC亚群CD8α-CD11c+细胞。将20只BALB/c小鼠随机均分为4组,A组为过继转移日本血吸虫尾蚴感染鼠CD8α-CD11c+型DC并诱发过敏性哮喘组,B组为过继转移正常鼠CD8α-CD11c+型DC并诱发过敏性哮喘组,C组为单纯过敏性哮喘组,D组为生理盐水对照组。A、B组小鼠经尾静脉过继转移5×105个DC,1h后A、B、C3组均开始诱发哮喘。4周后处死小鼠,取小鼠肺组织进行病理学检查。结果:肺部病理学结果显示A组小鼠肺组织炎症明显减轻,支气管管壁基本光滑,支气管内无黏液分泌。B组、C组肺组织炎症反应严重,支气管管壁增厚,管内存在大量黏液。D组小鼠肺组织无炎症反应。病理评分结果A组高于D组,低于B、C组,差异有统计学意义(P<0.05或P<0.01),B组和C组的差异无统计学意义(P>0.05)。结论:日本血吸虫尾蚴感染鼠CD8α-CD11c+型DC可明显抑制过敏性哮喘的发生。Objective: To study the inhibitory effect of CD8α^- CD11c^+.dendritic ceils (DC) on allergic asthma in schistosoma japonicum-infected mice. Methods: CD8α^- CD11c^+ DC from schistosoma japonicum-infected mice and naive were isolated and purified using microbeads respectively. Twenty BALB/c mice were divided into four groups randomly. CD8α^- CD11c^+ DCs (5 × 10^5) from schistosoma japonicum-infected mice were injected into the tail vein of mice in group A.CD8α^- CD11c^+ DCs (5×10^5) of naive were injected into the tail vein of mice in group B. Group C included simple allergic asthma rats. Rats in group D were given normal saline. One hour after injection, mice were sensitized and challengedwith ovalbumin to induce asthma in groups A, B and C. After four weeks, all mice were killed, and left lungs were collected for histopathological analysis. Results: Pathology results showed that pulmonary inflammation diminished significantly in mice of group A. There were more severe lung infection in groups B and C. No lung infection was found in group D. Results of lung pathological score was significantly higher in group A than that of group D, but significantly lower than that of groups B and C (P〈 0.05 or P〈 0.01). There was no significant difference between group B and group C (P〉 0.05). Conclusion: CD8α^- CD11c^+ DC can significantly inhibit allergic asthma in schistosoma japonicum infected mice.
