摘 要:目的探讨体外青藤碱增强骨髓间充质干细胞(BMMSCs)免疫调节能力的机制。方法原代培养小鼠MSCs。将青藤碱和(或)小鼠BMMSCs加入混合淋巴细胞反应中。第3天,以MTT比色法检测各组T淋巴细胞增殖情况。酶联免疫吸附试验(ELISA法)检测上清液中Th1细胞因子[白细胞介素2(IL-2)、干扰素-γ(IFN-γ)]和Th2细胞因子[IL-10、肿瘤坏死因子β(TGF-β)]的水平。结果 90%培养出小鼠BMMSCs细胞为CD29+、CD44+、CD45-、CD34-,是典型的MSCs。青藤碱联合BMMSCs组的吸光度(A)值为0.316±0.066,低于青藤碱组、BMMSCs组和阳性对照组的0.499±0.089、0.602±0.101和0.922±0.113,差异有统计学意义(P<0.05)。青藤碱联合BMMSCs组IL-2、IFN-γ水平分别为0.066±0.009和0.769±0.074,低于青藤碱组(0.259±0.017、1.023±0.095)、BMMSCs组(0.177±0.045、1.422±0.106)和阳性对照组(0.631±0.021、3.158±0.315),差异有统计学意义(P<0.05);青藤碱联合BMMSCs组IL-10和TGF-β水平为2.216±0.247和1.863±0.221,高于青藤碱组(1.437±0.072、1.112±0.095)、BMMSCs组(1.386±0.169、1.128±0.107)和阳性对照组(0.236±0.027、0.118±0.023),差异有统计学意义(P<0.05)。结论青藤碱能够促进小鼠BMMSCs的免疫调节作用,其作用机制可能与促进Th2细胞因子分泌以及抑制Th1细胞因子分泌有关。Objective To investigate the mechanism of sinomenine enhancing the immune modulatory effect of bone marrow mesenchymal stem cells(BMMSC) in vitro.Methods Primary mouse bone marrow mesenchymal stem cells were isolated.Sinomenine and/or BMMSC s were added to the mixed lymphocyte culture.On the third day,proliferations of T lymphocytes in each group were measured with MTT colorimetry.ELISA was used to detect the concentrations of Th1 cytokines(IL-2,IFN-γ)and Th2 cytokines(IL-10,TGF-β) in the supernatant.Results 90% of isolated mouse BMMSCs were CD29+,CD44+,CD45-,CD34-cells,which were typical BMMSCs.The A value of was 0.316±0.066 in the snomenine+BMMSCs group,which was lower than 0.499±0.089,0.602±0.101 and 0.922±0.113 in the sinomenine,BMMSCs and positive-control groups,respectively(P0.05).The concentrations of IL-2 and IFN-γ in the sinomenine+BMMSCs group were 0.066±0.009 and 0.769±0.074,which were lower than 0.259±0.017 and 1.023±0.095 in the sinomenine group,0.177±0.045 and 1.422±0.106 in the BMMSCs group,and 0.631±0.021 and 3.158±0.315 in the positive-control group(P0.05).The soncentrations of IL-10 and TGF-β in the sinomenine+BMMSCs group were 2.216±0.247 and 1.863±0.221,which were higher than 1.437±0.072 and 1.112±0.09 in the sinomenine group,1.386±0.169 and 1.128±0.107 in the BMMSCs group,and 0.236±0.027 and 0.118±0.023 in the positive-control group(P0.05).Conclusion Sinomenine could enhance the immune modulatory effect of mesenchymal stem cells.The mechanism of action may be related with promoting the secretion of Th2 cytokines and inhibiting the secretion of Th1 cytokines.
