摘 要:目的初步探讨宫内苯并(a)芘(BaP)暴露对胎儿癌症相关基因表达的影响。方法用高效液相色谱法检测20份新生儿脐带血血清中BaP的浓度,并按照BaP浓度将样本分成不同暴露组,用实时荧光定量PCR检测脐带血RPL27A(ribosomal protein L27a)、NUDT2(Nudix(nucleoside diphosphate linked moiety X)-type motif 2)和E2F1(E2F transcription factor 1)基因的表达,分析不同BaP暴露水平与癌症相关基因表达的关系。结果 20份脐带血血清BaP平均浓度为0.38±0.31μg/L。按照血清BaP浓度,将20份样本分成5个不同水平的暴露组。随着BaP暴露水平的增高,RPL27A和NUDT2呈现表达上调的趋势,而E2F1呈现表达下调的趋势。结论宫内BaP暴露可引起胎儿癌症相关基因表达的改变。Objectives To explore the impacts of prenatal benzo (a)pyrene (BaP) exposure on the expression of cancer-related gene of fetus. Methods BaP in cord serum of 20 newborns was detected by using high-performance liquid chromatography (HPLC)/fluorescence, and the mRNA of RPL27A, NUDT2 and E2F1 in cord serum was tested by real-time PCR. The relationship between BaP exposure level and gene expression was analyzed. Results The average level of BaP in 20 cord serum samples was 0. 38 ±0. 31 ug/L and the samples were grouped into 5 groups according to BaP levels. With the increase of BaP exposure, the expression of RPL27A and NUDT2 genes showed an up-regulated trend, while E2F1 gene showed a down-regulated trend. Conclusion Prenatal BaP exposure might induce a change of the expression of cancerrelated genes.
