摘  要:目的:建立Graves病小鼠模型,研究Graves病母鼠分娩后甲状腺自身抗体水平变化。方法:应用表达促甲状腺激素刺激激素受体(thyrotropic-stimulating hormone receptor,TSHR)A亚单位的重组腺病毒Ad-TSHR-289免疫雌性Balb/c小鼠制备Graves病动物模型。将6周Balb/c雌性小鼠随机分为实验组、对照组,分别用重组Ad-TSHR-289、Ad-β-gal经小鼠腿部肌肉注射,免疫剂量均为3.3×108pfu/50μl PBS,每隔3周免疫1次,共3次。免疫结束后,内眦静脉取血,检测血清甲状腺素(T4)、促甲状腺素受体抗体(TRAb),然后分别与Balb/c雄性小鼠配对,确定妊娠后继续饲养至分娩。分娩后1 d,处死母鼠,检测母鼠血清T4、TRAb、抗甲状腺过氧化物酶抗体(TPOAb)、甲状腺球蛋白抗体(TGAb),并分离母鼠甲状腺,行组织学检查。结果:实验组中96.7%的母鼠血清TRAb水平显著增高(P<0.05);实验组第3次免疫后有56.7%(17只)的小鼠达到甲亢标准,分娩后73.3%(22只)的母鼠达到甲亢标准;实验组中16.7%的母鼠血清TPOAb水平明显高于正常范围;母鼠血清TGAb较对照组显著增高,差异有统计学意义(P<0.05)。结论:利用Ad-TSHR-289免疫Balb/c小鼠可以成功制备Graves病小鼠模型。分娩后Graves病母鼠血清TPOAb及TGAb水平升高可能与TRAb有关。Objective:To illustrate the influence of "autoantibodies" in a post-partum period of a Graves' disease animal model. Methods:Female Balb/c mice aged 6 weeks were randomly divided into experimental group and control group,and injected intramuscularly with Ad-TSHR-289 (3.3 x 108 pfu/50 μl PBS) and control adenovirus (Ad-β-gal 3.3 x 108 pfu/50 μl PBS) respectively,three times at 3 weeks intervals.When the immunization was over,both groups were mated with male mice after getting a bit of serum from venae angularis for measurement of TRAb titer and T4 level. The mice were sacrificed 1 day after delivery, blood and thyroids were obtained. T4 level was detected by radio immunoassay. Levels of TRAb,TPOAb,TGAb were detected by eleetrochemilumineseenee immunoassay. The thyroids were used for histological examination. Results:TRAb positive rates were 96.7% in experiment group (P 〈 0.05). The incidence of hyperthyroidism in mice after delivery was 73.3% ,which was higher than that after the 3rd immunization (56.7%). TPOAb positive rates were 16.7% in experiment group. The titre of TGAb in experiment group increased siginificantly higher than that in control group (P 〈 0.05). Conclusion:An animal model of Graves' disease is successfully prepared by immunization with Ad-TSHR-289. The TRAb maybe involved in the increase of TPOAb and TGAb in postpartum mice with Graves' disease.
