摘  要:目的 探讨手术缝线携载c myc反义基因片断的可能性及可行性。 方法 保护薇乔缝线反复浸泡于c myc反义核酸溶液 72h ,溴化乙锭 (EB)标记后紫外灯下观察其吸附反义基因的情况。将吸附反义基因的缝线置于生理盐水中于不同时间点以紫外分光光度计测其吸光度 (A)值 ,绘出体外控释曲线。同时取释放液进行电泳分析。结果 吸附核酸的缝线经EB标记后在紫外灯下呈烧红的铁丝样 ,释放曲线提示 2h即达释放高峰 ,36h基本完全释放。电泳则可见随时间延长而逐渐变亮的电泳带。结论 保护薇乔缝线可较大量携载c myc反义基因片段 。Objective\ To investigate the possibility and feasibility of suture carrying c myc antisense oligonucleotide. Methods\ The suture (coated vicryl) was soaked in c myc antisense oligonucleotide solution repeatedly for 72 h. Then the suture carrying c myc antisense oligonucleotide was labeled with EB solution and observed under an ultraviolet lamp. The suture carrying antisense gene was emerged in normal saline and the value of absorbance ( A ) was measured by ultraviolet spectrophotometer at different time points. The releasing curve was made. Electrophoresis of the released solution was also performed. Results\ The suture carrying antisense oligonucleotide labeled by EB showed a reddish iron wire like. The releasing curve revealed that releasing peak reached at 2 h and the releasing was completed at 36 h. Electrophoresis indicated an increasing light band. Conclusion\ The Coated Vicryl can largely carry c myc antisense oligonucleotide which is released slowly.\;
