摘 要:[目的]掌握刺糖多孢菌生长代谢特性,为提高多杀菌素产量及发酵稳定性提供依据。[方法]对刺糖多孢菌斜面及种子生长规律、培养工艺及移种标准进行研究。[结果]确定斜面移种标准为平板单菌落移种第1代斜面,菌龄7 d;液体种子最佳培养工艺为添加30粒玻璃珠,温度30℃,初始pH值8.5;液体种子移种标准为生物量大于1.4 g/L、氨基氮75~80 mg/L、还原糖低于7 g/L、pH值低于7.5、种龄30~36 h。[结论]采用标准种子移种,多杀菌素产量提高58.5%,不同批次间产量差异小于5%。[Aims] The aim is to investigate the growth metabolism characteristic of Saccharopolyspora spinosa and to provide the basic materials for increasing spinosad production and fermentation stability. [Methods] The growth rule, cultivation process and inoculation standard of Saccharopolyspora spinosa were studied. [Results] The result showed that the cultivation of single colony on the plate for 7 days was the best inoculation standard of the slant strains. The optimized culture conditions of liquid seed were established as a combination of the glass bead of 30 grains in 500 mL shake flask, cultural temperature at 30 ~C and initial pH of 8.5. The inoculation standard of liquid seed consisted of biomass (greater than 1.4 g/L), amino nitrogen (75-80 mg/L), of reduced sugar (less than 7 g/L), pH (less than 7.5) and inoculum age (30- 36 h). [Conclusions] Employing the new inoculation standard, the spinosad production is increased by 58.5% and the difference of spinosad production among different batches is less than 5%.
