摘 要:以北京地区自然居群的大花杓兰蒴果为研究材料,对大花杓兰种子进行了非共生萌发研究。种子成熟度是影响大花杓兰种子萌发的关键因子:授粉后第6~8周,种皮呈透明至浅褐色,体式显微镜下可见白色球形胚时,种子萌发率较高;种子过于幼嫩或成熟均不易萌发。VWD改良培养基上种子萌发最好,萌发率可达68.06%。培养基中加入1.2 mg/L的KT可以促进种子萌发,并维持低褐化率,是适宜大花杓兰萌发的激素浓度。培养基中加入100 mL/L椰乳或100 g/L马铃薯有利于大花杓兰种子的萌发及生长。To maximize the in vitro seed germination of Cypripedium macrathos, the optimum time of seed collection,the suitability of culture medium, phytohormone and natural aqueous extracts were studied. The results showed that the seeds had higher germination rate when it was collected at 6 -8 weeks after pollina- tion, testa color was white to light brown, and white globular embryo could be seen under the stereomicroscope. The improved VWD culture medium was the best for seed germination. KT 1.2 mg/L could promoted seed germination,and retained low browning rate. 100 mL/L coconut milk or 100 g/L potato stimulated seed germination and growth.
